Liupao tea is a kind of dark tea, the material of Liupao tea was usded on this study, with3T3-L1adipocyte model and Insulin Resistance3T3-L1Adipocytes model to study the glucolipid metabolic effect. The results will provide theoretical basis of improving metabolic syndrome, weight-loss and lipid-lowering and adjusting the blood sugar for Liupao tea’s development. The main results were as follows:1. Effect of Liupao tea on proliferation and differentiation of3T3-L1preadipocyteDifferent concentrations of Liupao tea was used to study the effect on the cell proliferation of3T3-L1preadipocyte, detect the influence of cell proliferation by MTT method. Oil red O staining method, spectrophotometry analysis and RT-PCR were used to detect the effect of Liupao tea on cell differentiation. Research results show that Liupao tea do not exert effect on the growth and reproduction of3T3-L1preadipocyte. But the mRNA expression of PPAR gamma2and C/EBP alpha were decreased. Liupao tea significantly inhibit the differentiation of3T3-L1preadipocyte.2. Effect of Liupao tea on glucose and lipid metabolism in3T3-L1adipocytes3T3-L1preadipocyte can grow to undergo mature adipocyte cell line, round-shape and containing fat granule. Liupao tea can improve glucose uptake in3T3-L1adipocytes, and reduce the decomposition of adipocytes and release less free fatty acids. The expressions of HK、PFK-1、PK、GPAT mRNA in adipocytes was found to decreased. While the expression of CPT-I ACCm RNA significantly increased in groups treated with Liupao tea. Regulation of glucolipid metabolism maybe one of the mechanisms for Liupao tea to reduce weight and deter diabetes.3. Effect of Liupao Tea on Glucose and Lipid Metabolism in Palmitate-induced Insulin Resistance in3T3-L1AdipocytesInsulin resistance was induced in3T3-L1adipocytes after incubation in lmmol/L palmitate. The concentrations of glucose and NEFA decreased in medium. The mRNA expression of HK,PFK-1,PK,GLUT4,GPAT mRNA significantly increased in the Liupao tea groups. Meanwhile the expression of ACC、PTP1B mRNA was increased. High dose of Liupao tea(200μg/ml)raised CPT-I mRNA expression. Liupao tea had beneficial effect of improving insulin resistance induced by palmitate in3T3-L1adipocytes.
