The selection of matrix is necessary for the development of transdermal pharmaceutical products. Solvent acrylate pressure sensitive adhesives are commonly used matrix. They have many advantages, for example, transparent, good adhesive property, medium resistance and aging resistance performance, while adding other kinds of auxiliary material to the pressure sensitive adhesive, may result in poor cohesion, and thus, their application will be limited. Pressure sensitive adhesive should be modified to maintain its performance.The major objective of the research work was to improve the cohesion of pressure sensitive adhesive with organic metal and isocyanate crosslinking agent, the pressure sensitive adhesives contain carboxyl and hydroxyl groups, respectively.Duro TakB(DTB)pressure sensitive adhesive contains carboxyl groups, this study selected different kinds of organic metal crosslinking agent, we found that magnesium stearate has good compatibility with pressure sensitive adhesive,1% of magnesium stearate could meet the requirements of the adhesion performance of patch. We examined the effects of various components on the adhesion performance of pressure sensitive adhesive by orthogonal experiments.Duro TakA(DTA) pressure sensitive adhesive contains hydroxy groups,we used isocyanate crosslinking agent BAYMEDIX(?) A(BA) to crosslink with pressure sensitive adhesive. Moreover, the effects of concentration of crosslinking agent, type of catalyst, reaction temperature on the crosslinking degree of pressure sensitive adhesive were investigated as well. Crosslinking process were optimized by orthogonal experiment. And based on the small pilot study, pilot enlarge had been accomplished. The results showed that when the concentration of crosslinking agent was 0.4%, the reaction temperature was 110℃ and reaction time was 10 h. respectively. The final product could meet our requirement. The holding time was more than 24 h. Used in the existing patch prescription, The situation of poor cohesion was improved.Fourier transform infrared spectroscopy (FTIR), differential scanning calorimetry (DSC) and gel permeation chromatography (GPC), peel strength tester were adopted to characterize the properties of the DTA crosslinked pressure sensitive adhesive.
