Polysaccharides from Pleurotus ferulae was taken as the the research object to get such information :process conditions of Isolation and Purification,Applying infrared spectrum to analyse the structure of Pleurotus ferulae Polysaccharides(PFP),activity of scavenging free radicals and antibacterial,which can be used to provide basis for scientific research and industrial production after estimate the value of PFP.(1) Optimum condition was got by using experiment of single factor and response surface methodology : the extraction temperature was 60℃, the material-liquid ratio was 1:50(g/m L), time was 10 min and microwave power was 60 W.(2) It turned to be that the optimum conditions of removing protein from PFP was using Polyamide column chromatography. After explore the optimum process conditions of purification about PFP It got such conclusion : It was very effective when the content of polyamide was 3000 times of crude polysaccharides,and the polyamide column was eluted with 1.5 column volumes of deionized water,at flow rate of 1.5m L/min.(3) Analysis of PFP by infrared spectrum showed that Ultrasonic microwave assisted extraction(UMAE) had no effect basically on structure of PFP.(4) PFP had strong scavenging effects of DPPH radical, hydroxyl radical and superoxide anion radical. Scavenging effects had the positive correlation with the concentration. when concentration of PFP reached 5mg/m L, scavenging rate of DPPH radical, hydroxyl radical and superoxide anion radical was 67%, 63% and 59%,respectively.However,Antioxidant activity of PFP was weak compared with VC.(5) Polysaccharides from Pleurotus ferulae had no antimicrobial activity of Aspergillus niger and Saccharomyces cerevisiae, while It had antibacterial activities against Staphylococcus aureus, Escherichia coli and Bacillus subtilis, With minimum inhibitory concentration of 3.125 mg/m L,6.25 mg/m L and 3.125mg/m L,respectively.The antibacterial activity was positively correlated with polysaccharides concentration.
