Objective:To observe apoptosis of the rat myocardial cell resulted from fine particulate matter PM2.5) collected in the winter in Taiyuan,and to explore its mechanism.Methods:We chose fine particulate matter(PM2.5) collected in the winter in Taiyuan as the treated factors and rat myocardial cell as the treated object. MTT assay was used to detect cell vibility treated by different dose and different time by PM2.5.Fluorescence labeling probe and flow cytometry technique were used to measure the the ROS in the cell and the cell apoptosis.The Western Blot was used to test the expression of protein of treated after48 h by different doses PM2.5. Alpha View SA was used to analysis the stripe, Graphpad Prism5.0 was used to analysis data.Results:1. As compared with control, the cell vibility of different dose groups were(95.22±1.00)%、(89.50±0.56)%、(82.76±0.63)%、(73.53±1.02)%. The cell vibility of different time groups were(87.06±1.85)%、(74.57±2.46)%、(54.27±1.48)%、(47.26±1.82)%,The difference had statistical significance between groups(P<0.05).2. As compared with control,the yellow-green fluorescence was increased in treated groups,.The value(ROS) of treated groups was(12.54±0.31、23.27±0.46、33.48±0.66、46.09±0.32), as compared with contro( 8.53±0.23),the difference had statistical significance(P<0.05).3. As compared with control,the pueple fluorescence was increased in treated groups.The apoptosis rate of treated groups was(10.17±0.20)%、(16.47±0.19)%、(20.01±0.24)%、(29.27±0.31)%,as compared with control(5.07±0.15)%,the difference had statistical significance(P<0.05).4. As compared with control,the expression of bcl-2 protein was decreased, the expression of bax protein was increased, the expression of Caspase-3 protein was increased. The difference had statistical significance(P<0.05).Conclusion:The vibility of rat myocardial cell can be suppressed by PM2.5 with dose- effect relationship and time- effect relationship.The apoptosis of the rat myocardial cell resulted from PM2.5 may be associated with the generation of ROS,the decrease the expression of Bcl-2 and the increase the expression of Bax and Caspase-3.
