Shanxi vinegar vinegar is known as the "world a vinegar" reputation in the history of our country. Because of its unique fume fermented grains process, vinegar has a special color and smell, what make it become one of the spice of people love. Smoked and fermented grains technology of shanxi vinegar can promote maillard reaction activity into black melanoidin with a variety of functions,what make vinegar improve the anti-oxidation, fall blood pressure, fall hematic fat and bacteriostasis and other health care function. This experiment mainly choose the shanxi vinegar smoked vinegar fermented grains as experiment material, resulting from the fermented grains and the bacteriostatic test method of screening, can get the best method —— spectrophotometer method, quickly and easily detected the Bacteriostatic force of vinegar melanoidin. Vinegar fermented grains as raw materials, and then to vinegar melanoidin in extraction, purification, separation by laboratory ultrafiltration membrane separation device, the vinegar according to the size of molecular weight, get a different size of molecular weight of melanoidin components, and the various components of the enrichment and lyophilization, get the final powder. Choosed brush of shanxi vinegar antibacterial active ingredients and components, and then get the shanxi vinegar melanoidin bacteriostasis best components, and analyzes the component.The research content and main conclusions are the following:(1)In this study, spectrophotometry and microtitration were using to comp ara Vinegar melanoidins antibacterial activity. The results show that using sp ectrophotometry and microtitration assay cultured under nomial conditions af ter 24h growth of E. coli and containing different concentrations of E. coli fin e black vinegar OD values obtained after inhibition rate little difference betw een the two results and to analyze the difference was not significant, indicatin g that the measurement of the test bacteria culture OD value after a certain ti me, the two methods can be antibacterial. And when measuring the growth cu rve, the result of using microtitration method have large deviations, indicatin g that spectrophotometry are more stable than microtitration method. So next experiment we choose spectrophotometric study antibacterial properties. Vine gar melanoidins has antibacterial effects, and with the increasing of concentra tion of E. coli,the inhibition rate also increased, showing a dose-effect relatio nship. When the class of black sperm concentration 5mg/mL, can effectively inhibit the growth of bacteria, inhibition rate of 99%.(2)Melanoidins were separated by using ultrafiltration technology and then w e obtain different molecular weight melanoidins and analyzed the antibacteria 1 activity of different components,finally we find out the best one. UF with di fferent molecular weight will choose to separate melanoidins (less than 3kDa, 3-5kDa,5-10 kDa, greater than lOkDa), components of different molecular weight substances were detected antibacterial activity. Analysis results find o ut the 3-5K part have the greatest activity on the Suppression of inhibit Esche richia coli, Staphylococcus aureus and Bacillus subtilis antimicrobial. And wi th the increase 3-5kDa melanoidins concentration of bacterial inhibition rate also increased, showing a dose effect, and melanoidins minimum inhibitory c oncentration of 5mg/mL(3)Used sodium chloride to dissociate 3-5kDa melanoidin, and through the ultrafiltration, get the skeleton——PM and the dissociative content——BMC, using spectrophotometric method to do the bacteriostatic experiment, get the strongest bacteriostasis melanoidin compounds. The results show that the inhibition rate of PM is only 25.89%, compared with the same quality of 5 mg/mL M (93.11%) difference. Calculated to get that PM of melanoidin (3-5 kda) bacteriostatic force contribution is only 27.81%. The deduction is, the small molecules of melanoidin BMC may play a major role in bacteriostasis of melanoidin, is not melanoidin skeleton.
