This study was aimed at nicotine contents in the same kind of tobacco by different kinds method ,specially at the study is change of nicotine contents in tobacco after addition of Alone sample by HPLC process. Nicotine is primary component in tobacco by Utilizing GC-MS and nicotine contents are 6.50 mg/g by GC method with the recovery and coefficient of variation are 99.87% and 1.26%.The optimum conditions for extraction was proved practicable by condition filtration and orthogonal design with supersonic wave method .The results showed that the optimum conditions for extraction to ultrasonic method is : methanol (φ=40,20ml),and 15℃ for 30 min ,this paper has determined nicotine contents that is 23.50mg/g at absorption peak (λ=262nm) by HPLC with an eluent consisting of methanol –potass Phosphate alleciate (pH=3) with the recovery and coefficient of variation are 101.52% and 0.91%. The results of HPLC method are true and reliable and sensitive and the method save sample. Adding different breeds ,different places of Alone in tobacco ,in the light of ultrasonic method dealing with samples ,nicotine content was determined by assured HPLC method , The results showed that Alone can depress nicotine contents in tobacco ,all places are good to depressing nicotine in tobacoo, its peel of leaf and dry power are prominent .Leaf and peel of leaf of A.arborescens are notable. To sum up,Alone vera L. is more than A.arborescens. This paper systematically makes a series of experiments on the nicotine in tobacco,analyzing the received results more completely and scientific methods to the research on nicotine and the rational development of Alone . |
