A water soluble crude polysaccharide was extracted with hot waterand precipitated by alcohol from maize pollen. Protein was removed bySevag method and trichloroacetic acid (TCA) sedimentation respectively.The crude polysaccharide has been abstracted by the method of ethanolgrading. Eight fractions: PMAS/PMAT, PMBS/PMBT, PMCS/PMCTand PMDS/PMDT were obtained respectively. The efficiency ofextraction is 3.34%. No typical absorption of nucleic acid or protein wasobserved in UV spectrum. Infrared ray spectrum of PMCS indicates thatit has typical characteristic absorption peaks of polysaccharides. Thepollen polysaccharides of maize are white powder. HPCE (highperformance capillary electrophoresis) analysis indicated that theycontained glucose, arabinose and galactose.Then the PMCT was further purified with column chromatographyon Sepharose CL-6B. It was divided into three groups, named as PMCT1,PMCT2 and PMCT3. By gel filtration on Sephadex G-150 test, PMCT2was a single strait symmetry apex. The identification of purify impliesthat PMCT2 is respectively uniform on molecular weight. Its molecularweight estimated 4.88×104 dalton.The PMCS was chemically modified by chlorosulfonic axid-pyridine method. S-PMCS, a modified compound with 0.948 substituteddegrees was obtained. The IR spectrum of S-PMCS showed thatcharacteristic absorptions of sulfate ester bond were at wavenumber of1262.50cm-1 and 822.78cm-1.
