Photodynamic therapy (PDT) is a promising new treatment for light-accessible tumors. During the treatment procedure, the photosensitizer is a key factor, which has motivated lots of related studies for seeking sensitizers with good photodynamic properties. According to these reports, the amphiphilic metalphthalocyanine has received considerable attention. In this thesis, the DBU method was used to synthesize p-HPc, p-HPcCo, p-HPcNi, p-HPcZn. Through comparing their spectrum properties and the photodynamic properties, p-HPcZn was chosen for further study to investigate their interaction mechanism with protein or CT-DNA, as well as the photodamage ability. The results were shown as following:(1)Compared with unsubstituted Pc, the introduction of carboxyphenoxy improves the solubility of four Pc derivates, and can be attributed to amphiphilic phthalocyanine. This property is most important for photosensitizer to transmit in vivo and enrich on the surface of tumors.(2)The results of absorption and fluorescence spectroscopy showed that the central metal ions had effects on the spectral behaviors of these four Pc derivates. With the increase of the d orbit electron of the metal ion, their λmax red shifted. In these four Pc derivates, p-HPcZn was the best in absorption property and the strongest in emission intensity.(3)Because of the aggregation and the hydrogen band conformation influence, solvents had complex effects on absorption property of Pc derivates.(4)The experiments of photodynamic properties indicated that p-HPcZn possessed good photosensitive property among four Pc derivates, and can photodamage tumors through Type I and Type II mechanisms.(5)Through the research of the aggregration property, it can be known that, the stacking of p-HPcZn in ethanol-water solution aggrandized with the decrease of ethanol content, and the aggregation of p-HPcZn could be dispersed with the presence of cations and non-ion surfactants.
