The synthetic processes of two diagnosis drugs, fluoriscein sodium and indocyanine green, were studied in order to solve the problem of side effects caused by the impurities in two drugs and to offer high purity of diagnosis drugs for clinical uses.The components of domestic fluorescein sodium injection were analyzed by HPLC.The main impurities were separated and their structures were identified.The reasons of the impurities were investigated according to the chemical structures of impurities.The synthetic process of fluorescein sodium was improved and the reaction conditions were optimized.The contents of the impurities were reduced.The target compound was gained with a total yield of 64% and the purity more than 99.8%.The synthetic process of indocyanine green was studied.For the purpose of avoiding the side effects caused by sodium iodine residue, sodium acetate was used to replace of sodium iodine.The total yield was increased to 26% by optimizing the synthetic process.The structures of fluorescein sodium and indocyanine green were determined by 1H-NMR, 13C-NMR, IR and UV.
