Red Castor Seed Particle Preparation Process And Quality Standards

Objective: As the previous phase of gout, acute gouty arthritis (GA) will cause monstrosity of arthrosis and functional impediment in daily life; what is more, it will lead to serious nephropathy,cornary disease and even death. TCM plays an important role not only in diminishing inflammation,alleviating pain,regulating body immunity function,ameliorating syndrome and symptome, but also in promoting the drainage of uric acid,improving hemo-rheology and protecting joint. With the integration of TCM theory and updated pharmacological progress, this study,which will set a solid pharmaceutical basis for further research, mainly focused on a clinical empirical prescription and screened out the optimal extration and seperation technics by adopting modern filtration and design method.Methods: (1) Study on process technology: From the view of the content of paeoniflorin,diosgenin,arctinin,oleanolic acids the optimum extraction conditions were determined by single factor comparison design and orthogonal trials. With the application of central composite design, in terms of the content of total sapogenin and total polysaccharide, chitosan—one of the natural flocculants—was applied in the purification of Achyranthes bidentata water extract; and the optimal technics was also determined. Further more, the kinetics of flocculation and sedimentation process was preliminary studied. First, mix all the dry extracts, grind and sieve it; second, add some proper amount lactose, then use wet granulation method with adhesive, i.e. acrylic resin; Finaly, after desiccation of granules, the whole process is completed. (2) The study on quality specification: By using HPLC quantitative analysis on paeoniflorin has been made; By using TLC qualitative analysis on Paeonia Lactiflora Pall,Dioscorea spongiosa,Trachelospermum Jasminoides and bidentate achyranthes has been done.Conclusion: The effective materials of Chinese empirical prescription were extracted and made into granules. The study on quality specifications of the preparation were made to make sure the quality that the clinical medicine required for GA. It provides GA patients with a valuable medicine whose curative effect was confirmed and quality was controllable for the clinic. Meanwhile, this study established a solid pharmaceutical basis for future research.
