Xiaoer yanbian keli are complex extracts of Lonicera japonica, Platycodon grandiflorum A.DC,Pardanthus sinensis ker,Radix tinospoyae tinospora,Radix ophipogonis,Scrophularia ningpoensis, Dryobalanops camphor,Artificial oxgallstone ect.which having very prominent pharmaceutical function activity are widely used in clinical application.In this paper,aimed at resolving the lack of quality standard system of xiaoer yanbian keli on contents of compounds,safety,efficacy, and stability,a series of laboratorial studies on extraction process and quality control have been carried out.on the extraction process of xiaoer yanbian keli,according to the main composition and the physical chemical character of the prescription, the main compositions of Lonicera japonica Thunb Platycodon grandiflorum,Ophiopogon japonicus,Ker-Gawl,Belamcanda chinensis(L.)DC were analyzed for different areaes of production.In studying of extraction process using the yield of extract,the contents of the effective component as index,the best extraction process by water or aqueous alcohol were selected out by orthogonal design.The best extraction process by water was as follows:boiling time was 2,the first boiling was carried out with octuple water for 2h,and the second boiling was carried out with sixfold water for 1.5 h.The best extraction process by ethanol was carried out at 80℃for 2h twice.HPLC was used to determine Chlorogenic acid content.The operation was carried out in the Hypersil ODS chromatographic column (4.6mm×2.5mm×5μm)with the mobile phase consisting of a mixture of acetonitrile:0.4%phosphoric acid(10:90).The flow rate was at 0.8ml.min-1;UVdetection wavelength was at 327nm.The content of palanatine hydrochloride in xiaoer yanbian keli was analyzed by both HPLC and UV spectrophotometry.The operation was carried out in the Hypersil BOS chromatographic column(4.6mm×250mm×10μm)with the mobile phase consisting of a mixture of acetonitrile:water(containing 3.4g KH2PO4 and 1.7g CH3(CH2)11OSO3Na per 1000 ml(2:3).The flow rate was at 1.0ml.min-1;UVdetection wavelength was at 348nm. Conclusion:The methods have been proven to be feasible and convenient for the identification and to be accurate and sensitive for the result of the qualitative and quantitative analysis,the quality control was also reliable.
