Taxol is a type of the most effective anticancer drugs, especially for uterine cancer, ovarian cancer and breast cancer. The increasing clinical demands for active Taxol and the exhausted supply of natural Taxus, initial source of taxol, have spurred to produce these drugs or its substitute. This paper wants to track this question by entophyte fermentation based on the endosymbiosis theory.Firstly, entophytes, including 42 strains fungi and 77 strains bacteria, were isolated from the roots and stems of Taxus chinensis and the former were morphologically identified belonging to 18 genera, 6 families, 5 orders and 1 class. Moniliales, which includes 83.33% of the whole strains, was believed the dominant order. While Olpitrichum, Aspergillus, Trichoderma were believed the dominant genera, which may include 23.81%, 14.29%, 11.90% of the whole strains in respective. 4 strains fungi and 10 strains bacteria which could produce taxol were found by the TLC and HPLC.RS-4 and SPH-4 were identified belonging to and Trichoderma respectively. It is the first time to find endophyte producing taxol in Aspergillus. After strictly selection we choosed SPH-4 and ZMZ-7 as the objective strains.Secondly, we researched the factors which have an important effect on the mutation of microwave, and made the death curve,found that 300s was the best irradiation dose.After twice microwave irradiation.the production of taxol has increased nearly 2 times.we also found 120s is the best irradiation dose for ZMZ-7, the production of taxol is increased nearly 10 times by microwave irradiation,UV irradiation and X-ray irradiation.By a series of fermentation test of SPH-W2 the mutation strain,we found starch medium was the best medium, and the glucose and the peptone respectively the best carbon resource and the best nitrogen research. We also optimized the ferment technology and found that 28℃,PH7 was the optimum technologic parameter of SPH-W2.Conclusively, we offered some references and gived valuable advice for the consecutive research of screening the new resource and its substitute,which helped to optimize the fermentation technology.Meanwhile ,the finding of bacteria producing taxol also rectified the endosymbiosis theory, and make the good preparation for the further research, such as gene project,etc.
