This paper has two greatly parts, first part is by systematic investigation from 2007 to 2008, the diversity of insect community of jujube yard ecosystems was studied in the Xinjiang Pimo Reclamation Regions. Hoping for providing a theoretical basis and based information for Protecting the balance of ecological system and diversity of biodiversity for this Regions.System investigation was conducted and insect community structure and characteristic and its diversity were studied in jujube yards of Xinjiang Pimo Reclamation Regions. The result showed that there were 42 species of insects that belong to 12 orders (including Acarina) and 34 families in jujube yard. Insect population quantity growth and decay with the seasonal change has Very close relation in the jujube park. Because of that greatly parts of insects just started take place in April and May,The quantity isn't much.The jujube park and it around plant is grow quickly and the category is abundant. Mainly have Lepidoptera Diptera and Homoptera. Particularly the Lepidoptera is Mostly.Insect community richness of July August and September greater than April May and June. Show an upward trend.Present the trend of the ascension.Pass to the diversity of insect community of jujube yard ecosystems's analyze show that the old jujube yard and the young jujube yard have notability different, The size of the tree will influence the diversity of insect community.The diversity of insect community of the young jujube yard obviously big in the old jujube yard. So because of the size of the jujube yard, The diversity of insect community will appear different.The diversity of insect community of the jujube yard intercrop cotton and the jujube yard intercrop melon has notability different. Remove individual of species (such as Aphididae), individual quantity of the jujube yard intercrop melon big in the jujube yard intercrop cotton. The insect community of intercrop cotton jujube greater than the intercrop melon jujube.So because of the different intercrop of the jujube yard, Diversity of insect community will appear different.The second part clone Arginine Kinase of Tetranychus turkestani the cDNA encoding Homologous fragment. According to other insects of arginine kinase gene, designed a pair of degenerate oligonucleotide primers, using PCR, An approximately 503 bp of cDNA fragment was amplified, and then connect to the pMD18-T vector. Then, it was sequenced, the results show that Tetranychus turkestani AK gene have 503 bp. GLUSTAL X 2.0 software using the sequence alignment. Found turkestani AK gene 503bp and other insects AK genes were high conserved.We selected from the TTAK cDNA sequence contains the active site, building-specific dsRNA. Prepare for RNA interference technology.
