Preparation Of The Effective Components In Tripterygium Wilfordii Hook And Research On Quality Evaluation

Tripterygium wilfordii is one of traditional Chinese medicine and widely used in China.The exrtacts of T. wilfordii have various activities, inculding anti-inflammatory, anti-tumor, immunosuppressive and anti-HIV activities, etc. Its prepration was used for the treatment of rheumatoid arthritis, nephritis and other autoimmune diseases in clinical.T. wilfordii was reviewed in itsdistribution, application, chemical components, pharmacological function, clinica lapplications, etc.Further more, this review also covers the preparation of active compounds and detection of Tripterygium wilfordii.Several extraction and preparation of activities compounds from the extraction of twere studied.Moreover, the chromatographic finger print of its bioactive components was established by UPLC-PDA mehtod. The remarkable bioactive constiutents was identified by analysed some references compounds.HPLC method has been developed for the determination of triptolide; UV colorimetric method has been developed for the determination of total diterpene lactones tripterygium, different preparation methods from the preparation of the active ingredient of T. wilfordii. Using different extraction methods on total alkaloids of the preparation of the active ingredient of T. wilfordii by UV method the content analysis, which studide and compared the treatment of three samples of the preparation of active compounds.The result was that:First, Tripterygium exrtacts, CH3CL exrtacts and EtoAC exrtacts show biological effects in Immunosuppressive activity.Second,. MDS-gel was the best for the preparation of active compounds, and the content of triptolide total diterpene lactones tripterygium and total alkaloids the preparation of the active compounds was different.Third,10 remarkable bioactive constiutents was identified by analysed some references compounds, the contents of them were different.Fouth, seven compounds were isolated from original medical material of T. wilfordii by using a variety of chromatographic separation techniques (such as the preparation of high performance liquid chromatography) with the traditional extraction method of silica gel column chromatography. We also identified the structure of these seven compounds by assistance from analysis of various organic spectroscopy, including 1H-NMR,13C-NMR, HPLC-MS and high resolution mass spectrometry. Some of these were active compounds.In conculsion, the content of diverse of active ingredients of Tripterygium wilfordii Hook f and HPLC determination method were combimation to control the quality. In order to ensure effectivity and safety of the praeparatum of tripterygium wilfordii Hook f for the quality control of medical material provide theoretical basis.
