The production, breeding and protection of local goat breed(Funiu White Goat and Taihang Black Goat), the genetic polymorphisms of microsatellite markers and exon of BMP15 were reseached, their correlation to litter size and growth performance of two locals were analysised.Eleven microsatellite locus were selected of these, TGLA68, OarAE101, BM1329, LSCV043, BM143, OarHH55, GC101 were linked to the fecundity gene FecX and FecB mapped on chromosome 6 of sheep and cattle, BMS1248 located on chromosome 3 of cattle, MAF70 and OarHH35 located on chromosome 4 in sheep and BM1227 located on chromosome 8 in cattle. The genetic diversity of microsatellite locus for Two Local Goats were analysised. The results were as follows: 175 alleles were detected in Two Local Goat Breeds and an average was 7.95. Microsatellite locus were rich polymorphism and could be used for the analysis of genetic diversity. The average polymorphism information content (PIC), average number of effective alleles (Ne) and average heterozygosity (h) at eleven microsatellite locus in Funiu White Goat were 5.6083, 0.7940 and 0.8171, respectively; while the values in Taihang Black Goat were 5.7022, 0.7886 and 0.8173 respectively. Results of using least square analysis to research the influence of microsatellite loci on litter size and growth performence of Two Goat Locals showed that the 110bp and 110/110bp at OarAE101, 150bp and 125/150bp at OarHH55, 147bp and 147/160bp at BM1329 had positive on the litter size in Funiu White Goat. Some relatively genotype at BMS1248, MAF70 had positive correlation to the growth performance (body weight, body height, body length ) of two breeds.Primers were designed to detect the polymorphisms on exon 2 of BMP15 in two local goat breeds by PCR-SSCP. The results showed that three genotypes (AA, AB and BB) were detected in Funiu White Goat, and their genotype frequency were 0.128, 0.776, 0.085, respectively; However, only AB and AA were in Taihang Black Goat and their genotype frequency were 0.070 and 0.930, respectively. Results of using least square analysis to research effects of exon 2 gene BMP15 on the litter size. showed that the Funiu does with genotype AB had 2,42 kids more than those with genotype AA and BB(P<0.05). The Taihang does with genotype AA had 2,42 kids more than these genotype AB(P>0.05). The sequencing revealed that four mutations(456T→G, 466C→G, 510C→T, 511T→G) in AB for Funiu White Goat, which resulted in amino acid substitution of V155G and S171P. No mutation was detected in Taihang Black Goat. These results preliminarily indicated that exon 2 of BMP15 gene is associated with prolificacy in Funiu White Goat.The study was designed as the first step in detecting genetic polymorphisms of microsatellites marker and exon 2 gene BMP15, their correlation to growth and prolific performance of Funiu White Goat and Taihang Black Goat was analysede, which would eventually provide useful information for the MAS program in goat production.
