Improved method for the DNA extraction of Laminaria sp. gametophyte was established on base of the CTAB and SDS/K~ DNA extraction procedures. The acquired DNA with high quality can be used in the DNA fingerprint analysis. The results of RAPD, ISSR and AFLP for the cell lines of Lam maria sp. gametophytes indicated that: 1) RAPD marker is valid for identifying Lam maria sp. gametophytes. Its DNA fingerprints, constructed with three primers OPC2O, OPD2O and OPD 15, can be used not only for identifying the 23 cell lines of Laminaria sp., but also for distinguishing male and female gametophyte within one cell line. 2) Without knowing other DNA fingerprint data for Laminaria sp., we confirmed the reliability and validity of RAPD results with the ISSR technique. It is proved to be an effectual way for distinguishing the 23 cell lines of Lam maria sp. This procedure can also be useful to exclude the possible contamination caused by prokaryotic organism. 3) AFLP data showed high polymorphism on genetic characters for the Laminaria sp. gametophyte, and it may be available to the gene linkage analysis with specific character. 4) Preliminary work of SCAR was carried out on the base of RAPD results; this laid the foundation to marker-assistant selection and breeding for Laminaria sp. 5) Internal transcribed spacer 1 (ITS 1) analysis for 3F, 5F, 1 2M cell lines were made. Together with other Lam maria sp. cell lines?ITS 1 data registered in Gene Bank, alignment was made according to the similar sequences. It proved that 3F, 5F, 12M cell lines are not within the scope of Laminaria sp. In conclusion, RAPD, AFLP and ISSR fingerprints are effective ways for identifying and evaluating the germplasm of Lam maria sp. gametophyte. These fingerprint techniques provided the ways for the study of the germplasm preservation and selection for Lam maria sp.
