Study on Mechanism of Immunodepression of Experemental Immunologic Stress Chickens Abstract: 200 two-week-old chickens were randomly divided into treatment (T) group and control(C). The treatment (T) groups were divided into four groups, T1, T2, T3, T4. The C group were intramuscularly injected with normal saline, the dosage was 0.5m1 per chicken, and T1 group chickens were vaccinated with Newcastle Disease IV Series by putting drop in noses and eyes. In order to build the model of immunologic stress. T2, T3, T4 groups were vaccinated with amounts of Newcastle Disease I Series by intramuscular injection, the dosages were 10,20,lO,doses per chicken respectively. Round about three days of inoculation, the T4 group was fed quickwell stress added into the drinking water.After 1, 3, 5, 10, 15, 21, day of inoculation, serum, immune oragans, were collected to detect the antibody(Ab) against ND by using P -microscopic method; and the CD4 CD8 T-cell subsets by immunohischemistry. Meanwhile, the cortex of thymus of T3 group were collected to detect the super-microscope structure of T-cell on 1,3,5 days. Three conclusions were reached: (1) After immunologic stress, the Ab against ND of T2, T3 groups kept declining from 1st to 5th day, then increased. There was significant difference with C group; the T1, T4 groups kept rising; and it was significantly higher than that of the C group on 15th day. (2)On the 1st day after immuologic stress, the T-cell nucleus in the cortex of thymus began to focuse slightly, the nucleus were out of shape on the 3rd day, the heterochromatin agglutinated into a ball; on the 5th day, the cell nucleus were concentrative, the mitochondrions were full of bubbles and holes, the heterochromatin was out of cell and the cell dissolued.(3)After immuologic stress, the number of the CD4 T-cell of T2,T3,T4 groups began to decrease from 3rd to 5th, there was significant difference with C group. Except for thymus from 10th day, it began to increase. But the number of the CD8 T-cell of T2, T3, T4 groups began to increase from 3rd to 5th, there was significant difference with C group. Except for thymus from 10th day, it began to decrease.
