Several kinds of Aquatic animals were analyzed in genetic variation, relationship between filial generation with parents and germplasm identification using random amplified polymorphic DNA(RAPD) markers:1 . Genetic variation of three wild stocks of Meretrix meretrix was analyzed using random amplified polymorphic DNA(RAPD) markers, 50 primers got amplified bands in total of 60 primers, amplified bands of 15 primers were selected to statistic similarity index and genetic distances.Genetic distances of intra-stock of Liaonin, Jiangsu and Guangxi were 0.169 + 0.034 , 0.117 0.032 and 0.108 + 0.031, respectively; and those of inter-stock of Liaonin- Jiangsu , Jiangsu-Guangxi and Guangxi-Liaonin were 0.222 0.036 , 0.209 0.043 and 0.316 0.047, respectively, the results show genetic distances of three stocks increased following distances increased. The marker of S396-900bp was owned by Jiangsu and Liaonin stocks, and marker of S433-250bp was owned by Jiangsu and Guangxi stocks, three stocks were indentified by this two markers.2. The genetic similarities of silver crucian carp (Carasssius auratus gibelid) (CAG), allogynogenetic silver crucian carp (ASCC), para-hybrids of allogynogenetic silver crucian carp and red common carp (PHAR) and red common carp (Cyprinus carpio) (CC) were analyzed using Amplified Fragment Length Polymorphism of DNA (RAPD). The numbers of polymorphic bands of CAG, ASCC and PHAR are similar each other, but significantly lower than that of CC and the genetic similarities of CAG, ASCC and PHAR are similar each other, but higher than that of CC, indicating that CC are more diverse as a normal diploid species than the other three triploids. Some PHAR specific bands were identified from those shared by some or all populations, suggesting that genetic recombination may have occurred among these 4kinds of fishes. The genetic similarities increase step by step from 0.274 of CC, to 0.286 between CC and ASCC and 0.321 between CC and PHAR. In contrast, the similarities decrease gradually from 0.932 of CAG to 0.914 between CAG and ASCC and to 0.749 between CAG and PHAR. Clustering analysis showed that CAG merges with ASCC first, then with PHAR and finally with CC. These findings imply that the genetic backgrounds of ASCC and PHAR have differentiated gradually from that of CAG with the increase of CC sperm stimulation times. Our results proved that CC sperm has integrated its genetic materials into ASCC and PHAR genomes when it stimulates the development of triploidy ova, causing the economic trait superiority of ASCC and PHAR.
