Traditional method of kelp seed-reaing is founded by its life history in the early sixties. We have been using this method from then. With the development of technology of cell engineering, gametophyte clone seed-rearing is a new technology. This paper mainly discusses some key technique such as multiplication culture of gametophyte cell line, seed-adhering and seed-reaing.The results show as follows:1, Multiplication culture of cell line:In jars of 20L, the inoculating density is 0.5g/L.Providing proper conditions, experiments of cell line growth were conducted. The formula,y=-0.1118+1.3108x+0.085x2-0.0006x3 (R2=0.9985),was gained by regression analysis and it can be used to estimate products of clone (y is fresh weight,x is time).The results of the consumption of nutrition indicate that when the density of clone is lower ,changing the cultural fluid once a week, N6ppm and P1ppm is enough; when the density of clone is higher, changing the cultural fluid once a week, N-10ppm and P-2ppm is sufficient.The instantaneous growth rate of clone is much higher in the condition of suspending culture than that in the condition of stilling culture.The growth speed of clone is different in various inoculating density. The experiment result indicates: Lower is inoculating density, faster the speed of growth is.The results of large-scaled culture show that 48g clone crushed by tissue disintegrator in the course of initial period and its inoculating density is 0.35g/L, a month later ,the clone is disparted directly and its density is 1.5g/L, the highest density of each jar is 41 Og, the growth speed mostly doubled per week.2, Seed-adhering: Before cell line adhering, the female gametophyte and male gametophyte were cultivated on short light-period after they were mixed. The result shows:The development time of the gametophyte is about 11 days and the clone fresh weight increases 40%. The study of the development of gametophyte on different ratioes of famale and male gametophyte is conducted.The result is similar under the ratio 1:1 and 2:1 .The germination time of sporophyte is late and the rate of germination of sporophyte is low when the ratio is over 5:1.Parthenogenesis begins to appears under the ratio 5:1.The spraying weight for each screen is 3 gram.The way of indirect spraying is easy and convenient and the time of sporophyte germination is consistent.3 , Seed-rearing:In the period of seed-rearing, adopt still water after adhesion, slightlyflowing water after 24 hours (superficial flow speed under 5cm/s) and normal flowing water after 72 hours ; start to wash screen at the pressure of 0.5kg/cm2 as seedling grows to 8-16 Rows cell, then increase gradually the strength and times of screen washing ; meanwhile adopt technical measures of conventional method. Results show:gametophyte adhesion rate is over 80%, seedling is strong and seedling quality reaches state standard of Class 1 seedling.In the period from seed-adhering to seedling of 8-16 Rows cell, speed of growth and development of seedling is 20 days faster than that of seedling cultured by normal technique.This article also makes statistical analysis to the main economic characters of sporophytes cultured by two seed-rearing methods.The result shows aberrance degree of sporophytes cultured by cell line technique is apparently less than that of sporophytes cultured by usual means.
