The liquit culture of Cordyceps Sinensis mycelium was studied by statistical methods. The contrast experiment results showed that,glucose and corn powder were the adequate multi-carbon sources;yeast extract and soya flour were the adequate mulit-nitrogen soures. Nutyitional ingredients and their adequate combinationins were investigated by orthogonal devices. The optimum ratio of the ingredients was studied by the central combination design.The result was proved by regression equation in theory and got the optimum nutyitional ingredients combination(%):glucose 1.23,corn power 1.5,soya flour 2.0,yeast extract 0.57,malt extract l.O.The ratio of C/N is 4.8:1.The effect of culture time, initial pH, ethanol thicknesses, secondary supplementing sugar and inoculum quantity factors on biomass and polysaccharides of Cordyceps sinensis were exploited,the results showed the optimum culture time was 6d, initial pH was 5.5, ethanol thicknesses was 1.0%,and secondary supplementing sugar was Glu+Man+Gal(3:4:2)1.2g/100ml, inoculum quantity 5%,and the maximal output of polysaccharides and the mycelium weight reached 5949.6mg/L and 17.35g/L respectively.The method of adding some Chinese medicines extracts to Cordyceps Sinensis basic medium was adopted to select the kinds and that can stimulate the growth of Cordyceps Sinensis and the production of the Cordycepic polysaccharides by the standards of the weight of mycelium and the production of polysaccharides,in order to provide the fittest kinds of Chinese medicine prescription to ferment producting polysaccharides.In the meantime,the characteristic of adding some Chinese medicines extracts fermentation and its technique of Cordyceps Sinensis beverage were studied,the optimum technique parameters and fermentation mature conditions were determined by orthogonal test. The restraining bacterium effect of the Fermentative Fluid of Cordyceps sinensis was also studied,the results showed: the fermentative fluid of Cordyceps sinensis had a stronger restraining bacterium effect and a resisting heat ability.The crude polysaccharides of the mycelium and the fermentative fluid were respectively washing with distilled water and 0.1M,0.3M,0.5M NaCL solution by DEAE-32 cellulose column chromatography.The results were showed,their litmusless polysaccharides have the similar molecular weight and content; also their acidic polysaccharides also have the similar law.The characteristic peaks can be a standard of judging of Cordyceps sinensis polysaccharides.The experiments of grading trend deposit showed that receiving ratio had the increased trend as the ethanol thicknesses increasing; their polysaccharides contant had markedly changes;at the ethanol thicknesses of 30% and 70%,its had the maximum .It indicated that their polysaccharides molecular weight were pockety.The content of the components of polysaccharides was also mensurated by and stepping ethanol deposit.
