The polysaccharide content of Inonotus obliquus is obviously higher than most of the other large fungus. The most composition is glucan and some heteropolysaccharide which compose by glucose and a few other monosaccharides such as xylose. The pharmacological actions of polysaccharide content of Inonotus obliquus are antitumor, antiviral, scavenging free radicals, inhibition of lipid peroxidation generated, enhance immunity, etc.Inonotus obliquus, is a kind of perennial parasitic fungi and valuable medicinal fungus. Inonotus obliquus have complicated composition, such as polysaccharide, folic acid derivatives, aromatic fragrance oxalic acid, alkaloids and other active material. Inonotus obliquus polymyxin, mykol, lanosterol, ergosterin, triterpene compounds, aromatic acid and folic acid derivatives have been extracted. The research shows that Inonotus obliquus are wildly used to prevent and treat various diseases, have the effecting in anticancer, antiviral and resistance to the cardiovascular system diseaseThe research of submerged culture of mycelia in Inonotus obliquus in this paper and have culture medium optimization for polysaccharide which from physical activity Inonotus obliquus. Polysaccharide extracting is used the methods of reduced pressure concentration technology and alcohol precipitation for mycelia of Inonotus obliquus. The main results are showed as follows:The nutrients of culture medium for submerged culture of mycelia in Inonotus obliquus are screening by factorial experiment. The multivariate linear regression analysis of seven main factors, such as carbon source, nitrogen source and inorganic salt shows that the biggest factors of Inonotus obliquus growth are carbon source, nitrogen source, K2HPO4and MgSO4. So the optimization formula of culture medium for submerged culture of mycelia in Inonotus obliquus are3%corn flour,1.5%soybean flour,0.15%K2HPO4and0.1%MgSO4.The OD values in490nm of Inonotus obliquus fermented liquid are tested by phenol sulfuric acid method to determine the polysaccharide content of extracellular and intracellular. The polysaccharide content in intracellular is higher than extracellular after submerged culture of mycelia. The yield and purity of polysaccharide increased gradually with the extraction temperature rise. Polysaccharide can effectively dissolving and improve yield and purity rate because the penetration ability and solubility increased as the extraction temperature rise and the molecular kinetic energy increased. But the yield and purity of polysaccharide stabilized when the extraction temperature rose to80℃above. The yield and purity of polysaccharide also increased gradually with the extracting time increased. The yield of polysaccharide is growing faster in early period with large driving force because the polysaccharide concentration in solution is low, and big differs of concentration inside and outside of Inonotus obliquus cells. The yield and purity of polysaccharide stabilized as extracting time continue to increase. The possible cause is the polysaccharide concentration in solution heighten and the concentration difference become small as the extracting time extend, so the diffusion speed slow down and arrive the balance.The action, which polysaccharide diffused from Inonotus obliquus cell to dissolvent, is a diffusion process of concentration difference promotion. The large the driving force, the more polysaccharide diffused. But the cell membrane has retardation in polysaccharide diffused. So the more solvent consumption could increase the concentration difference and promote extracted polysaccharide but no influence on cell membrane. Higher dissolvent content has not big impact in yield rate of polysaccharide, and no increase in contents of polysaccharide. More dissolvent content is against the separation of polysaccharide concentration. So the best extraction technology of polysaccharide extracting is extraction temperature of80℃, extracting time of3.5h, dissolvent content of1:15.
