Effect Of Yinzhihuang Injection On Infectious Serositis Of Ducks By Artificial Infection And Its Mechanism

Yinzhihuang injection, a complex preparation of traditional Chinese Medicine, composed with Virgate Wormwood Herb, Cape Jasmine Fruit, Honeysuckle Flower extract and baicalin (account 4％), had displayed a wide range of pharmacological effect including heat-clearing and detoxicating, promoting urination and protecting liver. General all purpose to therapy hepatochlic hygropyrexia, nausea and vomiting, yellowish or reddish urine, active and chronic hepatitis in clinic. Contemporary research showed that YZH had the function of antibiosis, dephlogisticate and so on. In order to guide its application in clinic scientifically, we investigated the effect of YZH on infectious serositis of ducks by artificial infection and its mechanism.At first, we erected patho-model of infectious serositis of ducks. After conteracting RA 4 to 5 hours, the appetite of ducks began to decrease and they became depressed. Liver, pericardium and spleen began to appear affection 36h later. Typical affection such as cellulose cardiopericarditis, hepatic capsulitis and capsulitis by dissecting after conteracting RA 48h later. Survival rate of model group is 25％, YZH group is 75％after two weeks, compared with model group, P＜0.05.Next, we erected three patho-model: mouse auricle swelling induced by dimethyl benzene, rat swelling of toe induced by albumen and mouse weight of granulation tissue induced by agar. So as to estimate the effect of YZH on active and chronic inflammation. At last, effect of YZH on mouse auricle swelling is differential at different concentration, 4％YZH group is optimal, inhibition rate of swelling is 35.21％, 2％NASA group inhibition rate between 1％and 2％YZH group, inhibition rate of swelling is 12.68％. Effect of YZH on rat swelling of toe induced by albumen, 4％and 2％YZH group are better, inhibition rate of swelling are 22.30％and 21.03％, 2％NASA and 1％YZH group are feeble. 4％YZH group is optimal to the mouse weight of granulation tissue induced by agar, inhibition rate of swelling is 22.39％, 2％NASA group is worse than it. The results showed that YZH had great effect on active and chronic inflammation.In order to observe the antioxidation of YZH, we mensurated the activities of SOD, NOS, iNOS and contents of MDA, NO. The results showed that it significantly ameliorated macroscopic and histological damage, evidently elevated the activity of SOD, reduced the activities of NOS, iNOS and the contents of MDA, NO.Because of severe inflammatory reaction produced considerable NO, it damaged the membrane of cell and cell organ to induce mediators of inflammation liberation and generate inflammation. In this study, we found that YZH inhibited the formation of NO in the macrophage in a dose-dependent manner and time-dependent manner compared with control group.In conclusion, we concluded that the probable mechanism of YZH ameliorated inflammatory injury in RA-induced infectious serositis of ducks by its modulation of inflammatory mediators and antioxidation.
