Rhodiola,a wild plant of perennial herbage,is sub-rosebush of Rhodiola.L category of Crassulaceae section in rosebush item.It is officinal,edible and has the function of caducity staving,anti-hypoxia, radiationprotection,anti-fatigue and antivirus,etc.Pharmacological researches show that salidroside is the primary active ingredient of Rhodiola.L.Recently,with the gradual strengthening of the awareness of hazards of excessive UV radiation,development of sunscreen cosmetics has been concerned increasingly.Natural plants with non-toxic,no side effects have become the development trend of sunscreen.Salidroside has been extracted respectively by three different methods:reflux extraction, accelerated solvent extraction and ultrasonic extraction,with the best conditions of these methods determined HPLC.On the basis,the SCGE system triggered by UV using lymphocytes of white mouse has been established in order to detect the sunscreen efficacy of salidroside.The main contents and test results are as follows:In this paper,salidroside,which could be separated with acetonitrile and water(4:96),is measured by HPLC.Salidroside has been extracted respectively by three methods,with the best conditions of these methods detennined by orthogonal tests.Based on the comparison,it has shown that ASE is the optimum method to extract salidroside.The results of stability of salidroside indicate that salidroside is very stable under the conditions:irradiated by UV;preserved at room temperature or frozen for 10 days or kept in the non-strong alkaline solution,but it is easy to decompose under high temperature or in the alkali solution.Therefore,the best preservation conditions are that it should be kept in dark,cold and neutral solution.Electrophoretic conditions in SCGE,which are used to detect the role of radiation protection of salidroside,have been determined on the basis of establishment of model of UV mutagenesis.The safe use of the concentration of salidroside is 1mg·mL-1,or it has the poisoning effect on the cells.The study shows that salidroside has obvious protective effect on DNA damage induced by ultraviolet radiation,but it can not be higher than the safe use of concentration,otherwise salidroside will aggravate the injury.At the same time,it also has a clear protective effect on DNA damage caused by ethanol.Further study shows that other components of Rhodiola have a synergy effect with salidroside,in turn which enhances the protective effects of radiation injuries.
