This experiment use Rhodiola seed and leaf as explants, use MS as basic medium, study of the effect of their callus induction, growth, differentiation with different nitrogen sources, carbon sources, pH value and the medium hardness, at the same time rooting of regenerated seedlings, transplanting conditions were studied, the results show that:(1)Ratio of different nitrogen sources effect on callus induction of Rhodiola. Use different explants with different nitrogen content, use seeds as explants, the rate of callus induction were best with ammonium nitrate and potassium nitrate content for the 1000/2550 mg/L. With leaf explants, the rate of callus induction were best with ammonium nitrate and potassium nitrate content for the 500/3050 mg/L.(2) Different types of carbon sources effect on callus induction of Rhodiola, the best rate of callus induction medium with carbon source, followed by glucose, and finally the sugar.(3) The highest rate of induced with Rhodiola seed sucrose content of 30g/L; male and female plants leaves required different sucrose concentration. The highest rate of callus induction of female plant leaves were 30g/L; male plant leaf were30-70g/L.(4)The best value of Rhodiola seed callus range were pH 5.8~7.0; Rhodiola female plant leaf were pH 7.0, the most male plants leaf were pH 5.0.(5)Use Rhodiola seeds as explants, induction rate were not significant with different agar; use Rhodiola leaf as explants, the agar content of 7-10g/L help contribute to callus induction.(6) radiation length 14h, the light intensity 1000Lux, temperature 24℃, were good for callus induction.(7) The callus growth were fastest with ammonium nitrate and potassium nitrate 1400/2100mg/L in, sucrose concentration of 50g/L, pH value 5.8~6.2 in favor growth of callus.(8)The differentiation rate were highest of Rhodiola seed callus induced with ammonium nitrate and potassium nitrate content in 1000/1900 mg/L,1000/2550 mg/L; glucose medium for the differentiation rate of carbon source; the most suitable callus pH value of 6.0; sucrose concentration range of 10~30g/L; agar in 6~9g/L were good callus differentiation.(9) The best rooting medium were MS as Rhodiola seeds induced the regenerated plants. The regenerated plants hardened for 2-4days, room temperature 20~24℃, light length in the 12h/d, the most suitable soil is humus transplant:moss (1:1).
