Objective To investigate the biological behavior of the carcinoma associated fibroblasts (CAFs) of human gingiva carcinoma, including its regeneration, adherence and migration. To investigate the expression of its special correlated protein. To explore the relationship between the abnormal invasive ability of CAFs and the expressive level of fibroblast activation protein (FAP),α-smooth muscle actin (α-SMA) and matrix metalloprotease-2 (MMP-2).Methods The sample of CAFs was from a 42-year-old female patient suffering from gingiva carcinoma, and the sample of normal fibroblasts (NFs) was gained from the normal gum of the same patient. Both samples were cultured in vitro, researching on their biological disparity between the two purified strains by immunohistochemistry, proliferative activity detection, migration assay and transwell invasive test. Non-invasive stain (CAFs-NI) and hyper-invasive stain (CAFs-HI) were sieved from the CAFs by coated transwell caves. The expressive level of FAP,α-SMA and MMP-2 were analyzed and compared between the CAFs-NI and CAFs-HI by immunohistochemistrical staining, western immunoblotting detection and flow cell immunity counting.Results1 NFs and CAFs displayed a typical'S'morphous growth curve, with latency phase- logarithmic phase- platform. The proliferative activity of CAFs was super to that of NFs. The adherence rate of CAFs was higher than that of NFs. Scratching test showed that the CAFs posses a higher migration potential, repairing 0.8mm scratching by 24h. The invasive ability of CAFs was exceed that of NFs. (P<0.05).2 The positive expression of Vimentin and negative expression of Cytokeratin were shown in both CAFs and NFs, but FAP, MMP-2 andα-SMA were positive in CAFs while immunocytochemistrically negative in NFs.3 The expressive level of FAP,α-SMA and MMP-2 in CAFs-HI was super to That of CAFs-NI, according to immunohistochemistrical staining, western immunoblotting detection and flow cell immunity counting.Conclusions1 CAFs possesse higher ability of generation, invasion and adherence than NFs do in vitro.2 CAFs characteristically express FAP,α-SMA and MMP-2.3 The high invasive ability of CAFs has positive correlation with the expression level of FAP,α-SMA and MMP-2.
