Part IThe interactions between human tongue squamous cells cancer lines Tca-8113 and normal tongue mucoca fibroblastsObjective To study the interactions between human oral cancer cells and flbroblasts during oral tumor invasion and its mechanism to further explore the stroma roles in oral tumor invasion and metastasis .Methods To directly coculture human oral cancer cells with normal mucosa fibroblasts (NMFs) in vitro in order to minic oral cancer invasiveness. Two cells morphology and ultrostructures were observed under the normal microscope, inverted phase-contrast microscope and transmission electron microscope. Genes expression related to oral tumor carcinogenesis and progress was detected by semiquantitative RT-PCR before and after directly cocultured human oral cancer cells with NMFs.Results After coculture, two cells morphology were not clearly changed under the normal microscope. Fibroblasts, spindle-shaped with cytoplasmic processes, showed dentate nuclei and substantial rough endoplasmic reticulum, chondriosome and myofilament after cocultured with human oral cancer cells under...
