Study On Cantharidin Intervention In Acute Myeloid Leukemia Telomerase Reverse Transcrptase (htert) Methylation

Purpose:study the effect of the cantharidin, which is extracted from Chinese traditional medicine, for the methylation on telomerase reverse transcriptase (hTERT),to clarify whether there is demethylation treatment of the cantharidin on hTERT, explore the function of the cantharidin on treatment of acute myeloid leukemia.Method:adopt HL-60 cell line and inject the NOD/SCID rat body eradiated by 60Co 200cGy, forming NOD/SCID rat-human acute myeloid leukemia model, divided the mice into experimental and control group,and then the cantharidin is applied to disturb the experimental group.The RT-PCR Method and MSP Method are applied to observe the hTERT mRNA expression and hTERT methylation status of untreated group and experimental group respectively. Finally the results are to be analyzed. Results:the pathology on liver, spleen and thighbone of untreated group and experimental group showed the successful modeling; The cantharidin can apparently prolong the alive time (P<0.01) of the model rat with acute myeloid leukemia, and it is evident to influence the peripheral white blood cell of rat (P<0.01); The cantharidin can reduce the expression of hTERT mRNA (P<0.01); However, hTERT methylation exists on both untreated group and experimental group.Conclusion: The cantharidin can reduce the expression of hTERT mRNA, but it cannot produce apparent effect for the demethylation treatment on hTERT promoter methylation. Further study should be done by quantitative methylation examination.
