Objective:To investigate the difference of specific immune reaction between chronic obstructive pulmonary disease (COPD) model rats of qi-deficiency of the lung and that of qi-deficiency of the lung and kidney by detecting the concentration of IFN-γand IL-4 in serum and the expressions of CD4+T cells and CD8+T cells in the lung.Methods:30 healthy Wistar rats of SPF level were randomly divided into three groups including blank control group, qi-deficiency of the lung group and qi-deficiency of the lung and kidney group with 10 animals in each group. Passive smoking plus lipopolysaccharide (LPS) was used to establish COPD rat model of qi-deficiency of the lung. Passive smoking plus LPS plus subcutaneous injection of hydrocortisone sodium succinate (HSS) was used to establish COPD rat model of qi-deficiency of the lung and kidney. Then enzyme-linked immunoassay was applied to measure the concentration of IFN-y and IL-4 in serum, immunohistochemistry was used to observe the expression of CD4+T cells and CD8+T cells in the lungs and HE staining was used to detect histological changes of the lungs. Besides, ultrastructural changes in the lung were observed by electron microscope.Results:Enzyme-linked immunoassay showed that compared with normal control group, the concentration of IL-4 increased (P<0.01) and that of IFN-γdecreased (P<0.01) in other two groups, especially in qi-deficiency of the lung and kidney group(P<0.05, vs qi-deficiency of the lung group).HE staining showed certain inflammatory response exists in the lung of qi-deficiency of the lung group and qi-deficiency of the lung and kidney group. In these two groups, ciliated columnar epithelia fell off partially accompanied by significant hypertrophic response of goblet cells and glands.Immunohistochemistry demonstrated compared with blank control group, the expression of CD4+ T lymph cells decreased (P<0.05) in other two groups, especially in qi-deficiency of the lung and kidney group (P<0.05, vs qi-deficiency of the lung group); the expression of CD8+ T lymph cells increased in other two groups (P<0.05), but little difference existed between these two groups.Scan electron microscope (SEM) displayed bronchial morphological changes. Compared with blank control group, the mucosae of inner wall in the trachea were incomplete and epithelial cilia were collapsed and cohered, or became swollen, shorter and ill-arranged, even fell off completely accompanied by increasing number of goblet cells in local regions or with many adhered substances in qi-deficiency of the lung and kidney group. It is milder in qi-deficiency of the lung group.Conclusions: Compared with qi-deficiency of the lung group, severer immune disorder lies in qi-deficiency of the lung and kidney group, which can aggravate COPD. So it is inferred that immune disorder may be related with the pathomechanism of COPD from qi-deficiency of the lung to that of the lung and kidney. |
