Mahuang-fuzi-xixin decotion (MFXD) is a prescription from Shang Han Lun by Zhang, a famous doctor of Eastern Han Dynasty. It has been widely used to treat and prevent cold-induced febrile disease traditionally. MFXD is consisted of three herbs, Ephedra (Ephedra sinica Stapf, Mahuang); Asarum (Asarum heterotropoi Fr. Schmidt var. mandshuricum (Maxim.) Kitag., Xixin); Aconite Roots(Aconitum carmichaeli Dehx.; Fuzi), all of which are documented in the National Pharmacopoeia, this thesis is based on this priscription, and then to explore the activity of anit virus.The paper is composed by two parts below:1. The literature reviewThe paper summarized the progress of the clinical application of MFXD/the advance of macroporous resin/the update of serum pharmacochemistry of traditional Chinese medicine(TCM) and.the review of antivirus with TCM.2. the experimentThis part comprised 6 segments:(1) Gas Chromatography-Mass Spectrometry Analysis for Volatile Compounds in Mahuang-fuzi-xixin DecoctionTreated with steam distillation and headspace injection respectively, the volatile constituents in ephedrae herba of Inner Mongolia and Herba Asari of Jilin were analyzed by GC-MS. The above two sample preparation methods are focusing on volatile compounds with different boiling points; analyzing the herb with the above two methods is feasible to establish GC-MS representation system for ephedrae herba and Herba Asari.And then To analyze the volatile compounds in Mahuang-fuzi-xixin decoction by GC-MS, and then compared and analysed the volatile compounds from Ephedra and Herba Asari respectively. the result showes the volatile compounds of MFXD were mostly come from Herba Asari, this change maybe provide a basis for the compatibility mechanism and efficacy of MFXD.(2) The optimization of Extraction ProcessUsing high performance liquid chromatography (HPLC) fingerprint technology, factors of water amount, extraction time, extraction temperature and the number of extraction times that affect the yield of extract have been evaluated by orthogonal test design with multi-index of the content of ephedrine, hypaconitine, mesaconitine, the total area of characteristic peaks of HPLC and the yield of powdered extract. The optimum condition of the extraction was:water of 16 times the raw material amount at 90℃, twice for extraction and 1 hours each time.(3) Purification of MFXD by macroporous resinTo study the purification process with macroporous adsorption resin by the indicator of ephedra. To sieve the optimal purification of ephedrine in MFXD by exploring 4 types of macroporous adsorption resin. And D-101 macroporous adsorption resin can be used to purification of ephedrine in MFXD effectively among the resins.(4) The fingerprint of different parts of MFXD by alcohol washed and determination of alkaloids.According the propotion of priscription and the optimal extraction process, the MFXD is acquired, and then by the macroporous resin, we got the different part by the alcohol, with 30%,50%,70% and 95%. from the peaks of the number in the fingerprint and the content of Aconitum alkaloids and ephedrine, the fraction eluted mainly concentrated in 30% ethanol washed, and 30% ethanol eluation may be as a major research area.(5)The serum HPLC fingerprint of different parts of extraction of MFXDDoing experiment of serum pharmacochemistry with the 4 parts of MFXD eluted by macroporous resin, find that the group of 30% achieved the maxium srum drug concention at 30 minutes after administration, and got the less drug concention at 240 minutes; though the group of 50% achieved the maxium concention also at 30 minutes, then lowered at 180 minutes.(6) The antiviral activity of MFXDCollecting the essential oil of ephedra, asarum and MFXD, combined with the 4 different parts of MFXD eluted with alcohol by macroporous resin, then explore the antiviral activity of them, the findings demonstrated that the 50% and 70% ethanol eluation have the antiviral activity to RSV to some extent, the index of anti-virus both are 4, and the index of anti-FluA of 50% and 70% ethanol eluation are 2-4; the extraction of 30%,50%, 70% and 95% ethanol eluation have obvious anti-HSV activity, the index are 16,16,32 and 32 respectively.
