Objectives:To observe the effect of compound Modified SiNi Powder(JWSNS) on liver fibrosis in murine schistosomiasis formation.Methods:NIH rats (♀(?) half) were randomly divided into 5 groups:A group (control group); B group (model group); C group (praziquantel group); D group (praziquantel+JWSNS group); E group (praziquantel+colchicine group) In addition to normal mice, the mouse skin with each cover glass method, artificially infected with Schistosoma japonicum cercariae Article 26±2. Pathological liver tissue of mice after the formation of liver fibrosis, the mice infected 21 days after the beginning of treatment, D daily gavage group JWSNS decoction concentration 0.2 ml/per mouse/time (according to body equivalent surface area administered after conversion 0.0865g/times/20g mouse 2 times a day), E group fed with colchicine solution per day per rat 0.2ml/times/day, day 2, A, B, C Group saline gavage 0.2ml/20g. Treatment of mice in each group for 44 days, C group, D group, E group in the first 45 days after infection, respectively, to be administered praziquantel 300mg/kg for insecticide-treated one-time,3 days after the mice in each group continue to the original delivery. Treatment of the first 78 days of infection in mice detected by cardiac blood biochemical method serum alanine transferase (ALT) and plasma albumin (ALB) levels by RIA of serum typeⅢprocollagen peptide (PCⅢ) and transparent acid (HA) content; from liver tissue, spleen organ weight coefficient calculation; liver tissue hydroxyproline determination (HYp) content; liver tissue pathology observed changes in liver, detected by immunohistochemistry in mice liver tissue of collagen-Ⅰ,Ⅲcollagen; use of RT-PCR detected the expression of TGF-β1mRNA.Results:1. Organ coefficient (1) liver browser lowest coefficient of the normal control group, model group, praziquantel group JWSNS group, colchicine group were significantly higher than the normal control group (p<0.05); Modified JWSNS group was significantly lower than the model group, which is lower than praziquantel group, colchicine group (p<0.05). (2) spleen browser lowest coefficient of the normal control group, model group, praziquantel group JWSNS group, colchicine group were higher than the normal control group (p<0.05); JWSNS group was significantly lower than model treatment group, praziquantel group (p<0.05).2. Liver function (1) model group, the highest level of serum ALT, ALT JWSNS lower level group, compared with normal control group were statistically significant (p<0.05); JWSNS group lower levels of ALT, Compared with normal control group were statistically significant (p<0.05). (2) the model group significantly decreased serum ALB, ALB JWSNS group is higher than the model group, normal control group (p<0.05). 3. Serum HA, PCⅢand liver tissue HYp (1) HA was significantly higher than the level of the model group in each group (p<0.05); JWSNS group is higher than normal levels of HA, which is lower than colchicine group, praziquantel Group differences were statistically significant (p<0.05). (2) PCⅢlevel of the model group, praziquantel group was significantly higher than the normal control group (p<0.05); normal control group, between JWSNS more statistically significant (p <0.05), JWSNS group was lower than the model group, colchicine group, praziquantel group (p<0.05). (3) Hyp level of the model group, praziquantel group JWSNS group, colchicine group were significantly higher than the normal control group (p<0.05); the model group, praziquantel group, between colchicine Comparison of no statistical significance (p> 0.05); JWSNS HYp group were lower than the model group, praziquantel group, colchicine group, the difference was statistically significant (p<0.05).4. In each group I-type collagen,Ⅲcollagen (1) in liver tissue in each group of typeⅠcollagen average integral optical density comparison:normal control group was significantly lower than other groups (p<0.05); JWSNS Group, colchicine group the expression of typeⅠcollagen was significantly lower than the model group, praziquantel group (p<0.05); JWSNS colchicine group was lower than that group, compared the two groups were significantly different (p <0.05). (2) each group of typeⅢcollagen in liver tissue integral optical density mean comparison:normal control group was significantly lower than other groups (p<0.05); the model group, comparison between praziquantel group no significant difference (p> 0.05). Model group, liver tissue praziquantel group the expression of typeⅢcollagen than normal control group, JWSNS was significantly increased (p<0.05); the model group compared with the praziquantel group had no statistical significance (p> 0.05); JWSNS group, colchicine group the expression of collagen typeⅢwas significantly lower than the model group, praziquantel group (p<0.05); JWSNSⅢcollagen expression group was significantly lower than in colchicine group (p<0.05).5. Murine liver tissue of TGF-β1mRNA The mice liver expression of TGF-β1mRNA compared with normal mice have a statistically significant difference (p<0.05); the treatment group of mice the expression of hepatic TGF-β1mRNA than the model group (p<0.05); JWSNS mice the expression of hepatic TGF-β1mRNA than praziquantel group, colchicine group (p<0.05).Conclusion:1. Praziquantel against Schistosoma is the effects of drugs, mice infected with Schistosoma japonicum in the first 45 days simply give a one-time oral insecticide-treated, and can not stop the continuation of the progress of liver fibrosis.2.JWSNS medicine praziquantel combination can improve liver pathology of schistosomiasis in mice, reducing liver fibrosis in mice.
