Ovarian cancer is one of the three malignant tumors of the female reproductive system.Due to the lack of early effective diagnostic methods, furthermore operation and chemoradiation effects in a bad way,the 5-year survival rate of ovarian cancer patients is still low. With cerical cancer and endometrial cancer diagnosis and treatment progress, ovarian cancer has become a serious threat to the life of women malignant disease. So the problem we need to solve is that exploring early diagnostic methods and effective treatments, which becomes a grim task.Angiogenesis is a precondition of the tumor development, and many factors play roles in the formation of blood vessels. The new factors of angiogenesis, Eph receptor and their ephrin ligand, have been confirmed to have higher expression in many solid human tumors. They are belong toreceptor tyrosine kinases. The research of Eph/Ephrin in the ovarian cancer is less.ObjectieTo explore the expression of EphB4 and EphrinB in ovarian cancer and nomal ovarian tissues, so as to clear the contact between these two factors and ovarian cancer.MethodsWe used method of reverse transcription-poly-merase chain reaction(RT-PCR) to the expression of EphB4mRNA and EphrinB2mRNA in ovarian cancer and nomal ovarian tissues. To detect CD34 by immunohistchemical SP method, as micro vessel density count (Micro vessel density, MVD).Resultsâ‘ The relative content of EphB4mRNA was higher in the ovarian cancer group than in the normal ovarian group significantly,(p<0.05).The relative content of EphrinB2mRNA was higher in the ovarian cancer than that in the normal ovarian group significantly too(P<0.001).â‘¡The expression of EPhB4mRNA and EPhrinB2mRNA had a significant positive correlation with clinical stage and lymph node metastases (P<0.05).There was no relation to the patients'age, differentiation grade, pathological type, tumor size and ascites(P>0.05).â‘¢EphB4 and EphrinB2 may be closely associated with angiogenesis (P<0.05).ConclutionThe mRNA expressions of EphB4 and EphrinB2 in ovarian cancer were coincident, and positively correlated with MVD count. They play an important role in the oncogenesis and angiogenesis of ovarian cancer, and maybe do as an evaluation indexes of malignant disease. And may be a new breakthrough of ovarian anti-angiogenic treatment.
