The Establishion Of Rats Model With Phlegm Retention In The Lung Based On "spleen Is The Source Of Phlegm, And Lung Is The Storage Of Phlegm"

Objective:To duplicate rats model of phlegm retention in the Lung,we stimulated healthy rats with corresponding environment of laboratory,high-fat diet and fumigation,and then evaluated the model with related indicators,so as to deep explore and research the essence of a TCM theory,"Spleen is the source of phlegm,and Lung is the storage of phlegm ".Methods:48 healthy Wistar male rats were randomly divided into the following 6 groups:control group 2 and model group 4, placed in the corresponding conditions and stimulated differently at different stages.The changes of rats general conditions,especially oral and nasal secretions,cough,asthma and other specific symptoms were observed during the model making period.Specimens were collected on the 1st day,15th day,29th day and 34th day,and were observed their changes of blood lipids,blood rheology,trachea and lung tissue pathological, MUC (5AC), blood oxidative stress factor,phenol red testing expectoration and such indicators. SPSS16.0 statistical software was employed for statistical processing of experimental data and the analysis results.Results:The model rats physiologically have significantly audible airway sputum and coughing, wheezing, reverse hair, dry and dull body hair, extrados and curled sleeping, less movement, unresponsivement, eating little, thin stool, body weight loss and other symptoms. In histomorphology,ciliated columnar epithelial cells of trachea and bronchi partly exfoliating, goblet cell proliferating and secreting a lot, lung tissue obvious inflammatory exudation and hyperplasia can be seen easily.And in immunohistochemistry.the study showed MUCSAC secretion of model rats was higher than that of normal group; in phenol red expectoration experimental,model rats can be observed to secrete more compared with the normal group,significantly different; blood lipid and blood rheology changes indicators also increased significantly in model group rats.Conclusion:The indicators of pathology, immunohistochemistry, phenol red test and so on confirm that the model making is successful. The changes of physiology and blood lipids, blood rheology further reveal the relations of Spleen and Lung. And backward reasoning of the model making trial also further validates that " Spleen is the source of phlegm,and Lung is the storage of phlegm ",the classical theory of TCM...
