Objective:Observing expression of GCNF and Oct-4 in all malignant germ cell tumors in wax and P19 cells and its inducing cells, affording new methods to further study in pathogenesy,diagnosis and treatment of malignant germ cell tumors.Methods:1.Collecting and picking up all malignant germ cell tumors in wax that being kept on file in recent 30 years (1975.1-2007.12) by the pathology department of the first auxiliary hospital of the Shihezi university,making successive slice in 3μm, detecting expression of GCNF and Oct-4 by RT-PCR, Immunocytochemistry and double immunofluorescence technique at the level of mRNA and protein;2.Culturing P19 cells in vitro, inducing P19 cells with aMEM containing 8%FBS and 0.5umol/LATRA, observing the change of morpholage of cells, detecting expression of GCNF and Oct-4 in different terms with the methods above.Result:This study is the first time to detect GCNF is positive in all the maglinant germ cell tumors and normal testis tissues; Oct-4 expresses in most immature teratoma and all spermatocytoma and dysgerminoma, no expression of other sorts of malignant germ cell tumors and normal overy and testis tissues..GCNF and Oct-4 s stain location is in tumor cell nuclear, For the four kinds of maglinant germ cell tumors wich express both GCNF and Oct-4,the further differiation from aechaeo-germ cells, the more intensity of GCNF,the less intensity of Oct-4. Expressions of GCNF and Oct-4 in malignant germ cell tumors are in inverse correlations.After inducing P19 cell with ATRA,expression of GCNF ascends,while expression of Oct-4 descends. The expressions of GCNF and Oct-4 are in inverse correlations.Conclusion:(1) The positive location of GCNF in all malignant germ cell tumors hints that GCNF may be in correlation with the malignancy degree of tumor; (2) The dfferent expression of GCNF in all maglinant germ cell tumors hints that GCNF may be in correlation with the differentiation of tumors; (3) Expressions of GCNF and Oct-4 in malignant germ cell tumors are in inverse correlations hint that the function of GCNF in the malignant germ cell tumors may be acted by down-regulating the expression of Oct-4; (4) The variation expression of GCNF and Oct-4 in P19 and its cells induced hints that GCNF may be take part in the differentiation of the tumor cells by inhibiting Oct-4 gene.
