Objective: To evaluate the possibility of Fluorescence in Situ Hybridization(FISH) in detecting urothelial carcinoma of the urinary bladder. Methods: The probes of chromosome 17centromeres and chromosome 9 9p21 region (p16) were labeled by random primer method. FISH was performed on interphase nuclei of 80 urine specimens of cancer of urinary bladder ,20 normal donors were analyzed to establish threshold value. The pathological diagnosis was the"Golden standard", to analyse the sensitivity and specificity of chromosome aberration to diagnose Urothelial cancinoma of the urinary bladder. Results:The rate of aberration for chromosome 17 is 51.3% ,and 58.8% for chromosome 9p21 in urothelial carcinoma of the urinary bladder. The sensitivity is 86.5%,specifity is 100%. Suspected in 80 cases of bladder urothelial cancer patients, there are only 9 cases positive in the urine cytology, the sensitivity is about 12.2% (9/74). The nine cases are proved urothelial carcinoma of the urinary bladder by pathology, specificity is about 100% (6/6). The sensitivity of FISH is 85.2% and specificity is 81.6% in the diagnosis of hematuria in patients with bladder urothelial carcinoma. 17 chromsomes abbreviation have no significant correlation between pathological stage(p>0.05), but have significant correlation between pathological grade (p<0.01). 9 chromosomes abbreviation have no significant correlation between pathological stage and grade (p>0.05), Conclusion:FISH is believed to be a very important method in diagnosis of urothelial cancinoma of the urinary bladder and in the early detection of postoperative recurrence ,prognosis.
