| Objective: To established the modified rat orthotopic liver transplantation model of acute rejection, to observation the effect of immune tolerance induction in rat liver transplantation by pre-infusion of allogeneic lymphocytes treated by 5-Fu, and develop new ideas and feasible ways for induction of clinic transplantation immune tolerance under no common immunosuppression.Methods: Wistar and SD rats were used as liver transplantation donors and recipients, respectively and were divided into 4 groups as follows: group A, liver was transplanted from Wistar to SD; group B, recipients were pre-infused lymphocytes(5×106/ml)from Wistar rat 7 days and 4 days separately before transplantation; group C,recipients were pre-infused 5-Fu(7.5ug)treated lymphocytes(5×106/ml)from Wistar rat 7 days and 4 days separately before transplantation; and group D,recipients were pre-infused 5-Fu(15ug)treated lymphocytes(5×106/ml)from Wistar rat 7 days and 4 days separately before transplantation. Pathological changes and survival time of each groups were observed after liver transplantation.Results: Survival time of group A,B,C and D recipient rats was 6.7±1.5,10.3±2.7,19.0±5.3 and 11.7±3.9 days respectively.Group C by pre-infusion of lymphocytes(5×106/ml)treated by 5-Fu(7.5ug)were markedly longer than other groups(P<0.01);groupB,C and D were markedly longer than group A(P<0.01);group C were markedly longer than group B and D(P<0.01); group B and D have no difference(P>0.05). Pathological changes in group C were slighter than other groups.Conclusion: The modified two-cuff methods shorten the operation time in rat orthotopic liver transplantation model,and enhanced the stability and increased survival rate of rat orthotopic liver transplantation model.It can meet the requirements of experiments.Pre-infusion of lymphocytes treated by low level 5-Fu can induce immune tolerance in recipients after liver transplantation. |
