The Expression Of Cflip In Squamous Cell Carcinoma And Its Significane

Objective :This study aimed to investigate the role of celluar Fas-associated death domain-like interleukin-1β-conberting enzyme(FLICE)/caspase-8 inhibitory protein (cFLIP) in the pathogenesis of squamous cell carcinoma by observbing the expressions of cFLIP in the squamous cell carcinoma and its clinical significances.Methods : Immunoperoxidase staining method was applied to analyze the location of cFLIP expressions in 52 human squmaous cell carcinoma and 13 normal skin tissues.In addition, The expression of cFLIP messenger ribonucleic acid (mRNA) and protein was evaluated in the A431 cell line and Hacat cell lines.by RT-PCR and western-blot respectively.Results :The express rate of cFLIP is 67.31% in the experimental group and 15.38% in the control group, There was significant differences between normal skin tissues and squamous cell carcinoma(P<0.01).The expression of cFLIP is significantly associated with pathological grade and lymphnode invasion,There were no correlation between expression of cFLIP and age,sex and location of lesion(P>0.05) Furthermore, the expression cFLIP was positively correlated with the expression of PCNA(r=0.781,P<0.01). c-FLIP expression at either its mRNA or protein level was significantly higher in A431 than that in Hacat cell line(P<0.01).Conclusions : cFLIP is constitutively and more intensely expressed in SCC than that in normal tissues and cell line.The overexpression of cFLIP might be involved in the pathogenesis and the development of skin squamous cell carcinoma .
