Objective To investigate the effect of blockingβ-catenin by siRNA on expression of VEGF and MMP-9 and MVD in the nude mouse model of endometriosis, and whether the signaling wathway could accelerate the adhesion,invasion and angiogenesis of the implantation of the endometrium of endometriosis.Methods The female nude mouse model was developed by placing human endometrium fragments into the peritoneal cavity. Then, these eighteen had been surgeried nude mice were randomly and equally divided into RNAi group, negative control group and blank control group. Each animal was injected intraperitoneally and daily for 5days. After intraperitoneal injection of the last 24h, animals were sacrificed, collected and homogenized. RT-PCR and Immunohistochemistry were used to detect the expression ofβ-catenin,VEGF,mmp-9. The blood vessel density was estimated by counting staining with an endothelial marker CD34.Results the expressions of VEGF and MMP-9 mRNA and protein were down-regulated after gene silencing ofβ-catenin of the implantation of the endometrium of endometriosis in tne nude mouse model by RNAi (p<0.05). MVD in the RNAi group was less than the negative control group and blank control group (p<0.05).Conclusion siRNA-mediated inhibition of wnt/β-catenin limited not only the expression of VEGF and MMP-9 mRNA and protein, but also MVD. Wnt/β-catenin signaling pathway may activate implantation of the endometrium of endometriosis of the overall level.
