Objectives:To detect the expression of TGF-β1 and TGF-β1 signal transduction negative regulator Smad7 in prostate cancer by means of immunohistochemical streptavidin/peroxidase technique, and analyze the relations between the expression and the clinicopathologic characteristics, and discuss their function and significance in carcinogenesis,progression of prostate cancer,providing a approach for the diagnosis and treatment of the tumor.Methods:50 cases of prostate cancer (PCa) tissues and 25 cases of benign prostate hyperplasia (BPH) tissues were selected to detect the expression levels of TGF-β1 and Smad7 protein by means of immunohistochemical technique. Analyze the relations between the TGF-β1,Smad7 expression and the clinicopathologic characteristics, and then analyze the correlation of TGF-β1 and Smad7. All statisties analyzed by software of SPSS13.0.Results:1.The expression of TGF-β1(1)There were 35 positive cases in 50 samples of prostate cancer tissues, and the positive rate was 70.00%.In comparison with benign prostate hyperplasia tissues(24.00%,6/25) the difference was significant(P<0.01). (2)In prostate cancer group, the positive expression of TGF-β1 among the different pathological grading or different clinical staging was significant (P<0.05), and in those who had no bone metastasis was 59.38%, while it was 88.89% in those who had bone metastasis ( P<0.05).2.The expression of Smad7(1)There were 32 positive cases in 50 samples of prostate cancer tissues,and the positive rate was 64.00%.In comparison with benign prostate hyperplasia tissues (32.00%, 8/25) the difference was significant (P<0.01). (2)In prostate cancer group, the positive expression of Smad7 among the different clinical staging was significant(P<0.05),but not the pathological grading(P>0.05). The positive expression of Smad7 in those who had no bone metastasis was 56.25%, while it was 77.78% in those who had bone metastasis (P>0.05). 3. The correlation between the positive expression of TGF-β1 and Smad7 in prostate cancer: There was a significant positive correlation between TGF-β1 and Smad7.Conclusion:1. The difference of the positive expression of TGF-β1 and Smad7 between prostate cancer tissues and benign prostate hyperplasia tissues was significant, which maybe demonstrates that TGF-β1 and Smad7 participate in the course of carcinogenesis in prostate cancer.2. The positive expression of TGF-β1 among the different pathological grading or different clinical staging was significant,in prostate cancer, bone metastasis which maybe indicates that TGF-β1 is a promoting factor in the malignant progression, invasion and metastasis of prostate cancer. So the clinical detection of TGF-β1 in prostate cancer may contribute to the prognosis of prostate cancer, and can be an effective index to determine the pathological grading and clinical staging.3. The positive expression of Smad7 among the different clinical staging was significant, in prostate cancer, but not the pathological grading and bone metastasis, which maybe indicates that Smad7 take part in the course of progression, invasion and metastasis in prostate cancer. Smad7 is a phenotype of malignancy in prostate cancer and can not be an effective index to determine the pathology grading.4. The expression level of Smad7 showed increasing tendency with that of TGF-β1,and there was a significant positive correlation between TGF-β1 and Smad7,which indicates that Smad7 maybe take active effect in the carcinogenesis of prostate cancer by regulating the expression of TGF-β1.Therefore,we believe that TGF-β1 and Smad7 might take synergistic effect in the carcinogenesis, progression of prostate cancer, and the combination of them can be an effective index of the development and prognosis of prostate cancer.
