In recent years, it has been reported that the Eucommia male flowers have significant sedative and hypnotic effects. But whether the other parts of Eucommia plants have similar activities still not known. More works are needed on d the substances basis of sedative and hypnotic activities in Eucommia. In the current study, we research on the sedative and hypnotic activities in different parts of Eucommia (male flowers and leaves), and the sedative-hypnotic substances basic of Eucommia. That will provide theoretical basis for further research and product development on Eucommia.In this study, firstly, Different polarity active components in Eucommia were extracted with the system solvent extraction. Secondly, Primary active extracts were obtained through direct sedative and hypnotic experiments in mice, the synergy effects test with pentobarbital sodium, and anticonvulsant tests. Thirdly, we got the compounds form the active extracts through the chromatographic separation method and determine its chemical structure. Lastly, the activities of compounds have been determined in experiments in mice.The results showed that the different polarity extracts of Eucommia male flowers possess obvious sedative and hypnotic effects, the effects of ethyl acetate extracts. N-butanol extracts and total alkaloids are more noticeable than other extracts. The leaves and male flowers of Eucommia have similar sedative and hypnotic activities. And the active parts are also the ethyl acetate and n-butanol extracts in leaves of Eucommia. We obtained seven compounds form active extract of Eucommia leaves, and chose four compounds (DZY1, DZY2, DZY3, DZY5) for further active tests. Results showed that the DZY2, DZY5 could significantly increase the rate of sleeping mice and inhibit the spontaneous activity. The compounds also have synergy effects with sodium pentobarbital, could enhance the sleeping number of pentobarbital sodium subthreshold dose, prolong the sleeping time and shorten the sleep latency of pentobarbital sodium superthreshold dose. The DZY5 showed the strongest effects of the sedative and hypnotic. The compounds of DZY3 had a obvious function of the nervous excitement and showed the significantly dose-positive correlation within finite dose range. Overall results showed that: DZY2 and DZY5 are sedative-hypnotic active ingredients of Eucommia, and the DZY5 may be the primary active ingredient.
