Influences Of The Serum Containing Paidu Baoshen Decoction On Expression Of α- Sma And Ctgf In Human Renal Tubular Epithelial Cells Stimulated By Tgf-β1

Objective Renal tubulointerstitial fibrosis is the principal pathological process underlyling the progression of CKD (chronic kidney disease) to end stage renal failure. However, we still do not have effective therapies to stop the progression of renal fibrosis. In view of the grave situation, we need explore the antifibrotic approaches which are clinical available in Chinese medicinal herbs. To investigate the influences of the serum containing Paidu Baoshen Decoction on expression ofα- SMA (α-smooth muscle actin) and CTGF (connective tissue growth factor) in HKC (human renal tubular epithelial cell) stimulated by TGF-β1 (transforming growth factor beta-1), in order to probe Paidu Baoshen Decoction how to exert effective mechanism of renal protective action and to combat tubulointerstitial fibrosis during CKD.Methods Medicated rabbit serum and normal rabbit serum were prepared. Then HKCs were divided into 5 groups (TGF-β1 group, the low-dose medicated serun group, the moderate medicated serum group, the high-dose medicated serum group,Normal rabbit serum group). Examine the expression ofα-smooth muscle actin(α- SMA) mRNA by RT-PCR ,and Immunohistochemistry was used to detect the expression of connective tissue growth factor(CTGF) in the level of protein.Results After 48hours of the TGF-β1 stimulation, compared with the normal rabbit serum group, the expression ofα-SMA mRNA increased dramatically in the TGF-β1 group(p<0.01); the different dose medicated serun can suppress the expression ofα-SMA mRNA in HKCs exposed to TGF-β1.The inhibitory action of the high medicated serun group was more obviously than the low medicated serun group(p<0.01). Contrasted to TGF-β1 group(5.50±0.57), the normal rabbit serum group(0.50±1.00) was decreased in the level of CTGF protein(p<0.01);CTGF in different medicated serun groups(20%,10%, 5%)was2.50±0.577,3.50±1.00,5.00±0.82,respectively.Conclusions The medicated serum produces favorable effects on renal tubulointerstitial fibrosis may be related to restrain tubular epithelial-myofibroblast transdifferentiation.
