Bupleurum included in Chinese Pharmacopoeia 2005 is dry root of Bupleurum chinense DC.or Bupleurum scorzonerifolium Willd.from Umbelliferae plants.is one of traditional Chinese medicines in common used.The tast and property of the herb is bitter and slightly cold.Bupleurum root mainly medicine,but also in its stems and leaves,whole plant for medicinal parts,such as Jiangsu and most of the eastern part of Anhui,Bupleurum scorzonerifolium conventional spring harvest the whole plant with root,commonly known as "Spring Chaihu" medicine often alternative medicine in the clinical application of Bupleurum.However,the chemical composition of Spring Chaihu has yet to see research and reports,the Spring Chaihu has not fully feasible the quality of the evaluation criteria,this paper study a systematic research for the chemical composition of spring Chaihu to establish the Spring Chaihu quality assessment indicators,medication safety for clinical protection.The main contents are as following:1,Water vapor distillation was used to extract the volatile oil of different harvest time of Spring Chaihu.The results shows that the content of volatile oil was height from april to the middle ten days of may.This was broadly consistent with the traditional harvest time of Spring Chaihu.GC-MS was used to analyze the chemical components of the volatile oil.The volatile fatty acids in oil of different harvest time of Spring Chaihu are higher,especially in n-hexadecane with higher levels of acid can be used as the chemical characteristics constituents of volatile oil from the spring Chaihu.2,Study on the extraction technics of total flavonoids in Spring Chaihu.The monothetic design and orthogonal design were carded out for the extraction of flavonoids in Spring Chaihu,the optimal exctraction conditions were as follows: extracted 3 times with 15 times 70%alcohol at 60℃,1hour for each time.The ultraviolet- visible spectrophotometry was used to study flavonoids of different collection period and parts.The better harvest time of Spring Chaihu was from April to May and flavonoids content was that:leaf>whole plant>stem>root.The total flavonoids content was that:Bupleurum scorzonerifolium Willd aerial parts>Spring Chaihu>Bupleurum Pharmacopoeia.3,We establish the HPLC fingerprint of flavonoids of Bupleurum scorzoneri folium Willd aerial parts from different Habitats and Spring Chaihu with the technology of Chinese crude drug fingerprint.And most of common peaks were identified by HPLC-ESI- TOF MS.The results shows that Bupleurum scorzoneri folium Willd aerial part contains 14 common peaks and every common peaks were completely separated.Ten components are identified by the analysis of the MS spectra. Samples from different areas of the highest peaks No.3,5,8 of rutin,quercetin glycosides and quercetin,a total of three peaks representing a total area of 67%~78%, the characteristics can be used as the main features of Bupleurum scorzoneri folium Willd aerial parts.The Spring Chaihu received 12 common peaks,and eight components are identified by the analysis of the MS spectra.The HPLC fingerprint can be seen from the 4th peak for the Spring Chaihu flavonoids characteristic components,accounting for a total peak area of more than 50%.which peak on the 4th peak of quercetin-3-O-glucuronide.In different parts of the Spring Chaihu fingerprint analysis can be seen,the root,stem and leaf fingerprint of Spring Chaihu has a greater difference,the fingerprint of leaves and whole plant was higher degree of similarity.4,The first time that quercetin-3-glucuronide was isolated and identified from the Spring Chaihu.And establish a quantitative method of Spring Chaihu by HPLC. The calibration cure was linear in the range of 4.375~140μg·mL-1(r=0.9998),the average recovery was 98.43%,RSD 2.26%(n = 6).This method is simple,accurate and reliable.The results show that the content of quercetin-3-glucuronide has a greater difference in different harvest time of Spring Chaihu,with the highest concents in mid-April,and the content of different parts in the leave of Spring Chaihu. significantly higher than its stem and root.At the same time,also found that Bupleurum chinense DC.and Bupleurum scorzoneri folium Willd aerial parts hadn't quercetin-3-glucuronide.The results show that quercetin-3-glucuronide become the characteristic consituent of Spring Chaihu. 5,Both saikosaponin b2 and saikosaponin d had attained from Bupleurum chinense DC.and establish a quantitative method of saikosaponin d in Spring Chaihu by HPLC-UV.The calibration cure was linear in the range of 5.025~402μg·mL-1(r =0.9996),the average recovery was 98.90%,RSD 2.79%.This method is simple, accurate and reliable.The results show that the content of saikosaponin d has a greater difference in different harvest time of Spring Chaihu,the content of saikosaponin d was height from march to april,and the content of different parts:root>stem>leave.At the same time,also found that Bupleurum L.from china pharmacopoeia were 6~10 times with Spring Chaihu.
