There are more than 20 kinds species and varieties plantes of Vitaceae ampelopsis,which are rich in natural resources and widely distributed.In addition to A.japonica(Thunb.)Makino commonly used for traditional Chinese medicine,the vast majority is prescription.they are proved by pharmacology experiments that can promoting blood circulation,anti-inflammatory and stop bleeding etc.So it can be used to treatment of rheumatic arthralgia,osteomyelitis and swelling of unknown diseases.In this paper,Meicha is one of prescription which is used in Hubei enshi.It has Significant effect to treatment primary hypertension in clinically,because of its power which can reduce high blood pressure and high blood lipid.Its main effective chemical constituent is dihydromyricetin(3,5,7,3',4',5'-hexahydroxy-2,3-flavanonol,ampelopsin / dihydromyricetin),also known as the dihydromyricetin,ampelopsin,is an important flavonoids.With the flavone become into hot spots of active ingredients in natural plant,this chemical constituent have also been people's attention and concern.From the current situation on dihydromyricetin which is large exist in plants of Vitaceae ampelopsis,such as widely distributed in Guangdong,Guangxi,Yunnan,Hunan, Hubei,Jiangxi provinces and regions of China.Its exist in stems and leaves of young plants by 27%- 28%,the highest position up to 40 percent or more in A.grossedentata (Hand.-Mazz.)W.T.Wang;A.cantoniensis(Hook.et.Arn.)Planch.stems and leaves In the content of up to more than 25 percent.It is rare that the monomer content so high in the natural vegetation.Meicha as a prescription has been for many years,have a clear blood pressure-lowering effect,the application has accumulated a lot of valuable experience. However,the current system of Meicha and a comprehensive study of the development of new drugs are not carried out,greatly restrict the application of herbal medicine.In this paper,the related study on the chemical composition and fingerprint spectrum of Meicha has been done,to further its development into new drugs of Chinese medicine to provide adequate experimental basis. First,we collect some correlative data and literature of Meicha to determine Meicha's effective chemical ingredients,pharmacology effect and chemical quality, which point the way of how to design the technology of extraction.We isolated and purified the chemical constituents of A.megalophylla by solvent extraction and column chromatography.As a result,8 compounds were isolated and 5 compounds were identified by using physicochemical properties,spectral analysis (1H-NMR,13C-NMR,MS)and hydrolysis products.They wereβ-sitosterolⅡ, oleanolic acidⅢ,quercetinⅥ,myricetinⅦ,AmpelopsisⅧ,compoundsⅠ,Ⅳ,Ⅴstructure to be determined,of which compoundsⅡ,Ⅲ,Ⅵ3 components were first isolated from this plant.Secondly,the use of high-performance liquid chromatography to study on a fingerprint of Meicha.We investigated ten batches of Meicha with different harvest time.The HPLC standard fingerprints of the effective part of Meicha showing 12 common peaks,was analysed and established.By comparison of the retention time and the on-line UV spectra of chemical standards,peak 3,9 and 11 were identified as ampelosin,quercetin and myricetin,respectively.Use the HPLC chromatography to conduct the fingerprint.The technical parameters of the HPLC standard fingerprints study are consistent with national food and medicines Authority promulgated "Chinese medicine injectables fingerprint reader technology requirements (provisional)" related requirements.The analysis method has good credibility and stability.The fingerprint is able to evaluate its quality in a scientific and effective way. Thus the quality evaluation standard which connects international rails for this medicinal plant formulation has laid the foundation.
