The objective of this part was to study the potency of LBP and APS as adjuvant of inactivated influenza virus vaccine. The adjuvant effect on vaccine was tested by co-immunization i.m. or i.p. of mice with 300μg,500μg or 800μg LBP or with 300μg,500μg APS and 100μg aluminum plus vaccine at a dose of 0.015μg.Each mouse was administered twice with a 3-week interval.The results showed that,when co-injected by i.p.or i.m. with LBP or APS,the A/PR/8 inactivated influenza virus vaccine could strongly elicite a higher level of IgG in a dose-dependent manner than that of vaccine alone group.And the immune effects of 500μg,800μg LBP or 500μg APS were equal to aluminum group.The study of the effects of pachymaran as adjuvant of A/PR/8 inactivated influenza virus vaccine at a dose of 0.015μg or 1.5μg. The adjuvant effect of inactivated influenza virus vaccine was tested by co-immunization i.m. of mice with 200μg or 1000μg of pachymaran and 100μg of aluminum plus vaccine at a dose of 0.015μg or 1.5μg. At the same time,mice were inoculated i.m. with vaccine alone or PBS at the same doses.Detect the serum of mice for IgG or IgG1, IgG2a three weeks after immunization.Then mice were challenged with a lethal dosage (40×LD50) of A/PR/8 influenza virus Survival rate and body weight loss of mice were observed for three weeks.The results were as follows:(1)Enhanced immune effects of pachymaran to A/PR/8 vaccine at a low dose of 0.015μgThe serum IgG,IgG1,IgG2a of 1000 u g pachymaran group were greatly higher than those of 200μg pachymaran group or those of vaccine alone group,and gave mice lower weight loss but also enhanced survival rate,which is equal to aluminum.(2)Enhanced immune effects of pachymaran to A/PR/8 vaccine at a high dose of 1.5ugPachymaran can also argument the level of IgG,IgG1 and IgG2a,but weaker than aluminum group.After being challenged with a lethal dose of A/PR/8 influenza virus,pachymaran group got a similar weight loss to aluminum group.The objective of this part was to compare the different effects of MF59 in various routes.The comparative study was tested by co-immunization of mice once or twice with MF59 and A/PR/8 inactivated influenza vaccine through i.p.,i.m.,i.v. and i.n.. Serum specific antibodies were measured. The mice were challenged with A/PR/8 influenza virus at a lethal dose. The survival rate and body weight change were observed.The results showed that route i.p. was the best;i.m.,i.v. and i.n. were followed.
