Ursolic acid and Oleanolic acid are triterpenoid compound,they have anti-tumor,anti-inflammation, protect liver and beautiful white functions.In this paper we established HPLC determination and study on high effective separation and purification process of Ursolic acid and Oleanolic acid from waste residue of Catharanthus Roseus.Here are the main conclusions:1.Established the HPLC deterrmination method of Ursolic acid and Oleanolic acid.A HiQsil C18 column was used with the mobil phase methanol:acetonitrile:0.5%ammonium acetate=67:12:21(V:V:V) at the UV detection wavelength of 210nm,the flow rate was 1mL/min,the column temperature was room temperature.2.Established separation and purification process of Ursolic acid and Oleanolic acid.The detailed parameters are:(1) Aggrandizement extraction process was:using 90%ethanol as extractine solvent, sample and extraction solvent rate was 1:10,air flew rate was 0.2m3/h,extraction time was 25min,extraction times was 3 times.(2) 100.0g waste residue of Catharanthus roseus was extracted by 90%ethanol and got 19.0g product,the purity and recovery of Ursolic acid were 11.00%and 97.57%;the purity and recovery of Oleanolic acid were 3.01%and 99.72%.(3) Then the 90%ethanal product was extracted by ethyl acetate:using urtrasonic and stiring as extractine method,extractine times were 3 times.19.0g 90%ethanol product was extracted by ethyl acetate and got 4.84g product,the purity and recovery of Ursolic acid were 37.32%and 86.36%;the purity and recovery of Oleanolic acid were10.33%and 87.26%.(4) Then ethyl acetate product purification by silica gel column chromatography:using 300-400 mush gel silica as stationary phase,n-hexane:ethyl acetate=5:4 as mobile phase, sample quality was 0.2g/10g and flow rate was 3mL/min.4.84g ethyl acetate product purification by silica gel column chromatography and got 2.28g purifier product,the purity and recovery of Ursolic acid were 68.14%and 86.05%;the purity and recovery of Oleanolic acid were 17.79%and 81.21%.(5) Then Ursolic acid and Oleanolic acid were dissolved in ethanol,crystallized at 4℃. Then got 1.505g crystallization of Ursolic acid and Oleanolic acid,the purity and recovery of Ursolic acid were 74.15%and 76.39%;the purity and recovery of Oleanolic acid were 19.91% and 78.56%.(6) During the whole progress,the total recovery of Ursolic acid and Oleanolic acid were 55.39%and 55.53%.
