Rosa Multiflora Thunb belongs to the rosaceae plant genus Rosa, aka spend rose,is a kind of important as a food and medicine plant resources, rich nutrients, has the very high research value.The paper investigated the determination extraction and fingerprint of oleanolic acid(OA) and ursolic acid(UA) from Rosa Multiflora Thunb with the advanced method. The toxicological study of kunming mice.These researches laid on useful for studying Rosa Multiflora Thunb and functional product. The main results were as followed:1) Established a new analysis method for OA and UA presented in Rosa Multiflora Thunb by IIPLC:The content of OA and UA in different arts(roots, stems,fruits) of Rosa Multiflora Thunb have been compared,and taken analysis on content of OA and UA in Rosa Multiflora Thunb fruit from different origin by HPLC. The OA and UA was better separated within 20 minutes, it had good linear relationship between peak area mass concentration between 0.02468~0.2468 mg/m L and0.02523~0.2523 mg/mL with the correlation coefficient of 0.9994 and 0.9996 respectively. The recovery rates were 99.96%(RSD 0.74%) and 100.03%(RSD0.95%). The content of OA and UA in fruits reached the highest value. The Rosa Multiflora Thunb from Kuqa and Atux had the highest content of OA and UA, So that could be used as the origin for the research of Rosa Multiflora Thunb.2)Response Surface Optimization Extracting of OA and UA in Rosa Multiflora Thunb Fruits:Extract OA and UA in Rosa Multiflora Thunb fruits by ultrasonic extraction method, and take analysis on content of OA and UA by HPLC.Based on single factor experiments, response surface methology(RSM) was combined to optimize the process.The optimum conditions of OA was:the ratio to liquid 16:1mL/g,extraction temperature 41℃ and extraction time 37 min. UA that the ratio to liquid 17:1 mL/g, extraction temperature 40℃ and extraction time 38 min. At this conditions, actual yield of Rosa Multiflora Thunb fruits OA and UA were0.3981mg/g and 0.7170mg/g, it conforms to prediction results of the model(0.4067mg/g and 0.7311mg/g).The optimal extraction conditions are reasonable and easy to operate and the result could provide reference data for industrial production of extracting OA and UA in Rosa Multiflora Thunb fruits.3) Study on the Rosa Multiflora Thunb fruits in the HPLC fingerprints:To study the HPLC fingerprints in Rosa Multiflora Thunb fruits from different origins. Results shows that 7 peaks were observed on the HPLC fingerprint, but the contents werequite different. The precision, stability and repeatability were good. The method can provide a reference for quality control of ginseng fibrous root and lay the foundation for futhuer research.4)Total saponins in Rosa Multiflora Thunb toxicological studies:This study to evaluate Rosa Multiflora Thunb total saponins of toxicity, toxicity experiment was carried out on the Rosa Multiflora Thunb total saponins, concluded from the result of the experiment, the drug total saponins extract LD50 > 5 g/kg, nontoxic compounds.
