Objective:Malignant tumor is the present primary disease that endangers the people of the whole world. Its morbidity and mortality are increasing year by year now. Of all the malignant tumors, esophageal carcinoma is the most common .China has the highest morbidity and mortality. In China of all the number of the people that died of carcinoma , the gastric carcinoma is the first, and the number of the people that died of esophageal carcinoma is second which is one forth of all died people. Esophageal carcinoma not only deprives the people lives, it makes the patients suffered so much, stressed a lot and burdened heavily. As a result, it is a very important and urgent mission for the medical workers. Now the therapies for esophageal carcinoma are surgery, chemical therapy and radiotherapy. Their effects are not satisfied. Therefore the research that develops a new anti-esophageal carcinoma medical is rather important.In recent years, Cinese traditional drugs are more becoming a new focal point because its effect is across the board and its side reaction is little. Ginseng is one of the new developed Chinese traditional drugs. Ginseng is the delicacies of Chinese traditional drugs. Now the research shows it can delay aging , increase intelligence and increase the immunity of the body. Recently, more researches find it can inhibit the tumor cell grow, encourage the tumor cell apoptosis, inhibit the tumor cell metastasis and induce the tumor cell differentiate. As a result, Ginseng has become the focal point of the people . To assess the effect of 20(R) -Rg3 on the proliferation, apoptosis and cell cycle of esophageal carcinoma cell line EC-9706 cell and explore its role in the change of endogenous VEGF secreted by EC-9706 cells themselves.Methods:EC-9706 cell are cultured with three concentrations of 20(R) -Rg3 (50μmol/l,100μmol/l,200μmol/l), cell proliferation was detected by MTT method, cell cycle and apoptosis were detected with flow cytometry, and the expression of VEGF was detected by immunocytochemistry method after 24 h and 48 h.Results:By the microscope we can find the shape of EC-9706 cell is not regularity. The margin of cells becomes blunting and the photonasty is lower. The dead cells floating become more. It was found that Rg3 had significant inhibitory effect on the proliferation of EC-9706 cell after the cells being interfered with Rg3 at three different concentrations of 20(R) -Rg3 (50μmol/l,100μmol/l,200μmol/l). It is time-dependent and dose-dependent .The IC50 of 24h is 131.7μmol/l and the IC50 of 48 h is 81μmol/l. Rg3 could induce apoptosis and regulate cell cycle at three different concentrations of 20(R)-Rg3 after 48h. The apoptosis rates were 8.1%,9.7% and 13.8%, respectively. The control group had no apoptosis. Rg3 can suppress the expression of VEGF secreted by EC9706 cells themselves.Conclusion:With the action of 20 (R)-Rg3, the apoptosis of EC-9706 cell increased, the proliferation of cells was inhibited, and the level of VEGF secreted by EC9706 cell themselves declined. This might be one of the antiangiogenesis mechanisms of 20(R)-Rg3, and there appeared to be a dose dependent effect.
