Carcinoma of the breast is one of the most common malignant tumors in women. The incidence rate of breast carcinoma is the first or second place in some metropolises and medium-sized cities of our country.The therapy of breast carcinoma is comprehensive in current time.There are many methods,including operation treatment,chemical medication,radiotherapy,endocrine therapy,biological immunization therapy and so on.It is important to test estrogen receptor and progesterone receptor for the patients with carcinoma of the breast,because the result of the test can be very important prognostic factors and be a testimony of endocrine therapy.It has been indicated by substantial clinical information that the patients with estrogen receptor and progesterone receptor have a better prognosis than the negative ones.But the latter ones are between 30 and 40 per 100 patients with breast carcinoma.There are some studies showing that both the newly cloned BP1 gene and COX-2 are closely related with breast carcinoma.And there is some correlation between them and ER.Our experiment detected the BP1 and COX-2 mRNA expression by RT-PCR and used immunohistochemitry to detect BP1 and COX-2 in cells of breast carcinoma.We analysised the relationship between BP1,COX-2 and estrogen receptor.The correlation of BP1 and COX-2 was also studied.The aim of experiment is to better explain the pathogenesis of carcinoma of the breast and to provide more experimental evidence for clinical therapy of breast carcinoma. Methods1.We needed to culture two cell lines of carcinoma of the breast in vitro.They were the cell lines of MDA-MB-435 and MCF-7.2.Total RNA was extracted from the cells of breast carcinoma by using Trizol reagent according to the standard protocol.The quality of the RNA samples was determined by electrophoresis through agerose gels and staining with ethidium bromide.3.Total RNA were progressed directly to cDNA by reverse transcription using AMV reverse transcriptase.We also amplificated the objective genes to test the expression of objective genes by the technology of Polymerase Chain Reaction.4.We used immunohistochemitry to detect the expression of proteins BP1 and COX-2 in cell lines of breast carcinoma.5.Statistical analysis:t-test and Pearson correlation coefficient were adopted to analysis the experimental data at SPSS12.0.Results were considerable statistically significant at the 0.05 level.Results1.RT-PCR results indicated:The expression of mRNA of BP1 and COX-2 in cell line MDA-MB-435 was significant higher than that in cell line MCF-7.2.The same result was indicated in the study by the technology of immunohistochemistry,which was that the expression of the proteins of BP1 and COX-2 in cell line MDA-MB-435was significant higher than that in cell line MCF-7.3.There was a significant correlation between BP1 and COX-2.And analysis of correlation also indicated that there was a significant correlation among BP1, COX-2 and ER.Conclusion1.The expression of mRNA and proteins of BP1 and COX-2 in cell line MDA-MB-435 were significant higher than that in cell line MCF-7.2.The detection of the genes of BP1 and COX-2 in breast carcinoma can supply the evidence for the application of cyclooxygenase-2 inhibitor in the therapy of breast carcinoma,especially the ones that the estrogen receptor was negative.
