The Study Of Killing And Proapoptotic Effects Of Topoisomerase I Inhibitor On L1210/adm Cells

Objective:Observation and Study of Killing and Proapoptotic Effects of TopoisomeraseⅠ(topotecan) inhibitor on L1210/ADM cells.Method:The effect of topotecan on L1210/ADM cells was determined using MTT method, Annexin V FITC staining, DNA ladder electrophoresis and cell morphology analysis methods such as topotecan L1210/ADM of cells, apoptosis, the use of Caspase-8,3 inhibitor IETD-fmk, DEVD-CHO analysis of the topotecan-mediated destruction of the target cells, apoptosis and the relationship between Caspase.Results:TPT on L1210/ADM cell line has strong anti-role, and a clear time-dependent, at the same time, the target cells appear phosphatidylserine externalization, apoptotic bodies, such as the performance of the ladder electrophoresis. Caspase-specific enzyme inhibitor to join the group at the time of L1210/ADM cell lines were higher than the survival rate of single-use topotecan group (P<0.05), no performance-related apoptosis. Conclusion:Top otecan killing activity and was induce apoptosis to L1210/ADM cell lines, which could be dependeat on activation of Caspase-8 and Caspase-3.
