Objective: To make the bioactive IKVAVpeptide biomaterial self-assembled in vitro, and investigate its effect on neural progenitors cells (NPC).. Methods: 1. Synthesize the IKVAV peptide biomaterial: Dissolve the IKVAV-PA, control the key factors, and use the solution with bivalent cation to seduce the IKVAV self-assembled in vitro. Use the Transmission Electron Micoscope (TEM) to investigate the IKVAV petide biomaterial. 2. Get the hippocampusof the SD rat (borned of 1-3d) , use the method of non-bloodserum to getNPC. Use the method of immunoflourescence to appraise NPC. 3. Foster the NPC (the fourth generation) with IKVAV-PA and observe the adhesive and differentiation of NPC at 2h, 6h, 12h, 24h, then analyse the rate of the adhesive and differentiation. Use the statistics method to analyse the disparity with the control group (without IKVAV-PA) . Results: 1. The IKVAV peptide self-assembled to produce nanofiber gel successfully. 2. The IKVAV-PA ismakd to nanofiber with its diameter of 5. 0-9. 0nm, and formd a figrure of three-dimensional reticulation. It take on the length with number of count to hunders with a hardly shoring to the cells. 2. The NPC grew well whih IKVAV peptide, and the rate of adhesive and differentiation in the IKVAV peptide is higher than the NPC without the IKVAV peptide (P<0. 05) . It has the statistics sense. After 24 hours, the rate of adhesive of the NPCs in the IKVAV petide is 90. 7%, and almost all the cells had differentiated. And the control group only has the 10. 7% cells adhesived and 7. 7% cells differentiated. Conclusion: IKVAV-PA could self-assembled to produce nanofiber gel and promte the adhesive and differentiation of NPC.
