Preparation Of Polysaccharides From Vitis Viniferal L. Granules And Its Experimental Study On Immune Modulation Function

Objectives: To select the proper excipients for granules of Polysac- charides from Vitis Viniferal L.﹙VTP﹚and the preparative process, To establish the quality standard of Polysaccharides from Vitis Viniferal L﹙VTP﹚ Granules and study the two active compounds effects on the immune system of the immune suppressed mice. Methods:The present study included three parts of experiments.In the part 1,To find a better forming process of VTP Granule,With moisture absorption and building-up properties and tasty as the judgement criteria,the kinds and ratio of the excipients and process were sieved out.In the part 2 of experiments,Using phenol-sulfuric acid method to determine the content of VTP Granules.In the part 3,first,choosing eighty-four female BaLb/C mice,second,separating them into group for comparison, cyclophosphamide﹙CY﹚, with Lentinan pills,moderate dose of VTP Granules , substantive dose of VTP Granules with CY,moderate dose of VTP Granules with CY and minimum of VTP Granules with CY .third,separately dealing with them relevantly;finally,observing the thymus index and spleen index and activation of NK cells and transformation of lymphocytes of model mice of all groups.Results:﹙1﹚The optimal granulating process is:The ratio of VTP to lactose and mannitol was 1:0.375:0.125,The wetting agent was 65% alcohol .﹙2﹚The content of total astragalan in VTP Granules was 24.26%,the linearity of this method was very satisfactory﹙r=0.9998﹚,and the average recovery rate was 98.38%,RSD=0.85%.﹙3﹚Compared with the commonl group, the moderate dose group of VTP Granules can increase the spleen and thymus index of mice , activation of NK cells and the OD value of transformationof lymphocytes advanced markedly﹙P﹤0.01﹚,Compared with the model group,All immunosuppression groups have no significant effect on the thymus index of mice,but can increase the spleen index,comparing moderatedose of VTP Granules with common groups, activation of NK cells and the OD Value of transformation of lymphocytes advanced markedly﹙P﹤0.01﹚; comparing the treated group of substantive and moderate dose of VTP Granules with model group,activation of NK cells and the OD value of transformation of lymphocytes advanced markedly.Conclusions:The formula of granules is completed and reasonable,The moulding technology of granules is convenient and credible.The method of determination is simple, accurate and reproducible, and available for quality control of the granules. The granules can promote the immunological function in inmuosuppressed mice.
